In vitro characterization of the furin inhibitor MI-1851: Albumin binding, interaction with cytochrome P450 enzymes and cytotoxicity

Erzsebet Paszti-Gere et al.

Summary: The substrate-analog furin inhibitor MI-1851 can suppress the cleavage of SARS-CoV-2 spike protein and has a significant antiviral effect on infected human airway epithelial cells. MI-1851 does not interact with human serum albumin, but is significantly depleted by human microsomes. It does not inhibit CYP1A2, 2C9, 2C19, and 2D6 enzymes, but significantly suppresses CYP3A4 activity.

Interaction of the Emerging Mycotoxins Beauvericin, Cyclopiazonic Acid, and Sterigmatocystin with Human Serum Albumin

Eszter Fliszar-Nyul et al.

Summary: Beauvericin, cyclopiazonic acid, and sterigmatocystin are emerging mycotoxins that can contaminate food and animal feed, resulting in economic losses and health risks. Human serum albumin can form stable complexes with these mycotoxins, influencing their toxicokinetics and potentially serving as a cost-effective affinity protein. This study investigated the interactions of these mycotoxins with human serum albumin using various techniques, revealing the formation of low to moderately strong complexes. The interactions of cyclopiazonic acid with site markers suggest potential toxicological importance.

Probe into the Molecular Mechanism of Ibuprofen Interaction with Warfarin Bound to Human Serum Albumin in Comparison to Ascorbic and Salicylic Acids: Allosteric Inhibition of Anticoagulant Release

Hamid Reza Kalhor et al.

Summary: The study investigated the effect of commonly used drugs on the release of warfarin bound to HSA in silico. Both s-ibuprofen and ascorbic acid were found to exhibit allosteric effects on the release of warfarin from HSA, with ibuprofen showing a stabilizing effect and ascorbic acid aiding in the release through destabilization of HSA. Molecular dynamics simulations provided valuable insights into the interactions of drugs with the protein and the mechanism of anticoagulant release.

Interaction of SZV 1287, a novel oxime analgesic drug candidate, and its metabolites with serum albumin

Eszter Fliszar-Nyul et al.

Summary: SZV 1287 is a novel multi-target analgesic with promising potential for treating neuropathic pain. Studies have shown that SZV 1287 and its metabolites form stable complexes with albumin and exhibit activity in animal models. The interaction of SZV 1287 with albumin may play a crucial role in determining its pharmacokinetic properties.
