Role of Human Mesenchymal Stem Cells and Derived Extracellular Vesicles in Reducing Sensory Neuron Hyperexcitability and Pain Behaviors in Murine Osteoarthritis

Minji Ai et al.

Summary: This study aimed to determine whether mesenchymal stem/stromal cells (MSCs) and MSC-derived extracellular vesicles (MSC-EVs) reduce pain in patients with knee osteoarthritis (OA) by affecting sensory neuron excitability in OA joints. The results showed that mice treated with MSCs and MSC-EVs did not display pain-related behavior changes observed in untreated mice with OA, and MSC-EVs could prevent nerve growth factor-induced sensory neuron hyperexcitability. Therefore, MSCs and MSC-EVs may alleviate OA pain by directly acting on peripheral sensory neurons.

Mesenchymal stromal cell-based therapy for cartilage regeneration in knee osteoarthritis

Xiao-Na Xiang et al.

Summary: Osteoarthritis is a common degenerative disease that causes high socioeconomic costs and disability rates. The knee joint is most commonly affected, characterized by progressive destruction of articular cartilage, loss of extracellular matrix, and inflammation. MSC-based therapy has been explored as a new regenerative treatment for knee osteoarthritis. However, the detailed functions and mechanisms of MSC-based therapy, especially for cartilage regeneration, have not been explained.
