Disrupting Neurons and Glial Cells Oneness in the Brain-The Possible Causal Role of Herpes Simplex Virus Type 1 (HSV-1) in Alzheimer's Disease

Matylda Barbara Mielcarska et al.

Summary: Current data strongly indicate that herpes simplex virus type 1 (HSV-1) infection is a contributing factor to Alzheimer's disease (AD). HSV-1 infection causes damage to neurons and glial cells, as well as modifications in their environment, leading to impaired signal transmission and homeostatic and immune functions. This review discusses the pathological alterations in central nervous system (CNS) cells following HSV-1 infection, including inflammatory responses, oxidative damage, and cellular dysfunctions associated with AD development.

Diagnosis and Treatment of Sexually Transmitted Infections A Review

Susan Tuddenham et al.

Summary: Approximately 1 in 5 adults in the US had a sexually transmitted infection (STI) in 2018. Rates of gonorrhea, chlamydia, and syphilis in the US have increased, while rates of HSV-1 and HSV-2 have declined. Effective treatments are available for most STIs, but antimicrobial resistance limits oral therapies for gonorrhea and Mycoplasma genitalium, and no cure is available for genital herpes.

A combinatorial CRISPR-Cas12a attack on HIV DNA

Minghui Fan et al.

Summary: CRISPR-Cas12a is a gene editing tool that may have less off-target effects compared to Cas9. Researchers have found that using a combination of two crRNAs can effectively suppress HIV and prevent viral escape in long-term cultures. Interestingly, instead of excision, the inactivation of the HIV sequences was mainly caused by hypermutation.

Integrated Strategies for a Holistic View of Extracellular Vesicles

Valeria Rondelli et al.

Summary: Extracellular vesicles play a crucial role in cell-cell communication by spreading information, both beneficial and harmful. The complexity and heterogeneity of these vesicles make comprehensive studies challenging but necessary. This review highlights the effectiveness of studies that integrate diverse data on extracellular vesicles from different perspectives, aiming to improve our understanding of their clinical relevance. Integrative strategies combining biophysical approaches, omics technologies, and computational models will pave the way for new diagnostic, prognostic, and therapeutic applications.

SARS-CoV-2 infection and lytic reactivation of herpesviruses: A potential threat in the postpandemic era?

Jungang Chen et al.

Summary: The outbreak of SARS-CoV-2 has greatly stressed the healthcare system by causing the COVID-19 pandemic. In addition to respiratory and systemic symptoms, comorbidities such as chronic viral infections increase the risk of fatal outcomes. Recent reports have shown an increase in lytic reactivation of human herpesviruses in COVID-19 patients and vaccinated individuals. Coinfection, treatments, and vaccination for COVID-19 may aggravate herpesvirus-associated diseases by reactivating the latent viruses. This review summarizes recent clinical findings and limited mechanistic studies on the relationship between SARS-CoV-2 and human herpesviruses, highlighting the ongoing potential threat to human health in the postpandemic era.

Bartley P. Griffith et al.

Summary: A porcine-to-human heart transplantation was performed on a 57-year-old man with nonischemic cardiomyopathy who was not eligible for standard therapeutics, including traditional allograft. The patient was successfully weaned from ECMO and the xenograft functioned normally until sudden failure on day 49 post-transplantation. Further studies are needed to identify the mechanisms responsible for the changes observed in the xenograft.

Targeting herpes simplex virus with CRISPR-Cas9 cures herpetic stromal keratitis in mice

Di Yin et al.

Summary: The study targeted HSV-1 genomes directly using mRNA-carrying lentiviral particles (HELP), which efficiently blocked HSV-1 replication and reduced the occurrence of HSK in three different infection models. HELP was capable of eliminating the viral reservoir via retrograde transport from corneas to trigeminal ganglia, while inhibiting viral replication in human-derived corneas without causing off-target effects.

Development of Genome Editing Approaches against Herpes Simplex Virus Infections

Isadora Zhang et al.

Summary: Herpes simplex virus 1 (HSV-1) can cause cold sores or keratitis in healthy individuals, but severe complications in immune-compromised or neonatal patients. Current therapies can block viral replication but have limited effect on viral latency, necessitating the development of new strategies. Genome editing methods like homing endonucleases and CRISPR/Cas systems show promise in inhibiting HSV-1 infection.

Pathogenesis and virulence of herpes simplex virus

Shuyong Zhu et al.

Summary: HSV-1 and HSV-2 are two of the most prevalent human viruses worldwide, causing a variety of diseases. Immune responses play a key role in controlling HSV, but the virus has developed mechanisms to evade them. The severity of disease upon HSV infection varies among individuals, partly due to genetic polymorphisms. HSV has lytic and latent replication cycles, with the latter leading to disease.

Encounters between Cas9/dCas9 and G-Quadruplexes: Implications for Transcription Regulation and Cas9-Mediated DNA Cleavage

Mohammed Enamul Hoque et al.

Summary: Using dCas9, transcription levels of TH can be modulated by targeting different parts of a G-rich sequence within the promoter. The presence of GQs in target sequences impacts DNA cleavage activity of Cas9, with a significant reduction in cleavage activity when targeting high-stability GQs. Additionally, this reduction is more pronounced for the G-rich strand compared to the complementary C-rich strand.

Amenamevir, a Helicase-Primase Inhibitor, for the Optimal Treatment of Herpes Zoster

Kimiyasu Shiraki et al.

Summary: Acyclovir, valacyclovir, and famciclovir are commonly used for the treatment of herpes simplex virus and varicella-zoster virus infections. However, recent studies have shown that amenamevir, a helicase-primase inhibitor, has novel mechanisms of action against these viruses with promising clinical efficacy. Its antiviral activity is not influenced by the viral replication cycle and has been demonstrated to be non-inferior to valacyclovir in treating herpes zoster.
