The Therapeutic Potential of 2-{[4-(2-methoxyphenyl) piperazin-1-yl]alkyl}-1H-benzo[d]imidazoles as Ligands for Alpha1-Adrenergic Receptor - Comparative In Silico and In Vitro Study

Jelena Z. Penjisevic et al.

Summary: Adrenergic receptors, particularly the alpha 1-adrenergic receptors, have been extensively studied for their therapeutic potential in various disorders. In this study, compounds with potential alpha 1-adrenergic receptor antagonistic activity were identified through in silico docking, molecular dynamics simulations, and ADME calculations. These findings provide valuable insights for the development of novel drugs for neurological conditions.

MicroRNA miR-133a-3p Facilitates Adrenergic Proarrhythmic Ectopy in Rat Pulmonary Vein Myocardium by Increasing cAMP Content

V. S. Kuz'min et al.

Summary: Cardiac-specific microRNA miR-133a-3p modulates adrenergic signaling, leading to increased cAMP accumulation and enhanced intracellular beta-adrenergic signaling, which contributes to the occurrence of proarrhythmic ectopy.

A Concise and Useful Guide to Understand How Alpha1 Adrenoceptor Antagonists Work

Sidiane B. da Silva et al.

Summary: Adrenoceptors are receptors for catecholamines and play a role in regulating cellular responses. Alpha-adrenoceptors are used in the treatment of hypertension and benign prostatic hyperplasia, but their activation can also lead to adverse effects such as orthostatic hypotension. Understanding the structure-activity relationships can help in the development of drugs with improved selectivity and reduced side effects.

α1-adrenergic receptors accompanied by GATA4 expression are related to proarrhythmic conduction and automaticity in rat interatrial septum

Ksenia B. Pustovit et al.

Summary: The development of interatrial septum is a complex process mediated by alpha-adrenergic pathways. In normal conditions, the electrical activity in IAS tissue is less stable compared to myocardium. Stimulation of adrenoreceptors induces spontaneous action potentials in IAS tissue, which can be inhibited by alpha 1-blocker. Immunohistochemical data suggest the presence of GATA4 and abundant alpha 1A-adrenoreceptors in adult IAS myocytes, which may explain the ectopic automaticity of IAS myocardium.

The Antiarrhythmic and Hypotensive Effects of S-61 and S-73, the Pyrrolidin-2-one Derivatives with α1-Adrenolytic Properties

Klaudia Lustyk et al.

Summary: Heart rhythm abnormalities are a major cause of death worldwide, and current antiarrhythmic drugs have limited effectiveness. This study found that two novel pyrrolidin-2-one derivatives, S-61 and S-73, have potential antiarrhythmic properties and can protect against arrhythmias in rats.

Role of α1-blockers in the current management of hypertension

Hua Li et al.

Summary: There is increasing evidence supporting the safe use of alpha 1-blockers in the treatment of hypertension. These drugs are effective in controlling blood pressure in almost all hypertensive patients, with special indications including benign prostatic hyperplasia, resistant hypertension, and primary aldosteronism screen. However, careful consideration is required for the choice of long-acting alpha 1-blockers, potential orthostatic hypotension in the elderly, and fluid retention.

Klaudia Lustyk et al.

Summary: This study found that the novel compound HBK-10 has potent antiarrhythmic and hypotensive properties in rats, and it has a high affinity for alpha(1)-adrenoceptors. This suggests that 2-methoxyphenylpiperazine derivatives could be potential new drugs for the treatment of arrhythmia.

Attenuation of inward rectifier potassium current contributes to the α1-adrenergic receptor-induced proarrhythmicity in the caval vein myocardium

Alexandra D. Ivanova et al.

Summary: The study found that activation of alpha 1-adrenoreceptors in rat superior vena cava myocardium can lead to decreased conduction velocity, prolonged action potentials, reduced maximum upstroke velocity, and depolarization of resting membrane potential. These effects are mediated by the alpha 1A subtype of alpha-adrenoreceptors and may contribute to proarrhythmic conduction in the SVC myocardium.
