Evaluation of Neurofilament Light Chain as a Biomarker of Neurodegeneration in X-Linked Childhood Cerebral Adrenoleukodystrophy

Hongge Wang et al.

Summary: Cerebral adrenoleukodystrophy (CALD) is a devastating neuroinflammatory manifestation, and finding biomarkers for this disease burden has been a long-standing goal. This study suggests that plasma neurofilament light chain (NfL) could be an important indicator in evaluating CALD outcomes, with a significant correlation to cerebral spinal fluid levels and a potential for decrease after therapeutic intervention.

Lukasz Pawlinski et al.

Summary: This research aimed to identify new biomarkers and potential drug targets in Gaucher disease type 1 (GDt1) by analyzing serum proteins. A total of 31 proteins with significantly different concentrations between GDt1 patients and a control group were identified, mainly involved in regulating inflammatory processes and hemostasis. The study revealed new targets for further investigation that could be beneficial for the diagnosis, treatment, and monitoring of GDt1 patients.

Blood phenylalanine reduction reverses gene expression changes observed in a mouse model of phenylketonuria

Rachna Manek et al.

Summary: The treatment strategies for phenylketonuria (PKU) include PEG-PAL and rAAV-based PAH gene delivery, both of which lower Phe levels and have therapeutic effects on brain pathology; however, PAL delivery requires dose optimization, does not increase Tyr levels in the brain, and may elicit an immune response; the impact of high Phe levels on liver functions involves changes in lipid metabolism and upregulation of genes related to cholesterol biosynthesis.
