NLRP1 Inflammasome Activation in the Hippocampal Formation in Alzheimer's Disease: Correlation with Neuropathological Changes and Unbiasedly Estimated Neuronal Loss

Ena Spanic et al.

Summary: Neuroinflammation is a key pathological feature of Alzheimer's disease (AD). This study demonstrates that NLRP1 inflammasome is more active in the AD brain and is positively correlated with the number of neurofibrillary tangles (NFTs), suggesting a causal link between neuroinflammation and neurofibrillary degeneration.

Zinc Modulation of Neuronal Calcium Sensor Proteins: Three Modes of Interaction with Different Structural Outcomes

Viktoriia E. Baksheeva et al.

Summary: Neuronal calcium sensors (NCSs) can complement their calcium sensing activity by sensing zinc fluctuations. Different proteins in the NCS family have varying abilities to bind zinc ions, with some proteins showing conformational changes and regulation of dimeric state in the absence of calcium, while others become insensitive to zinc in the presence of calcium. Based on their zinc-binding properties, NCS proteins can be divided into physiological Ca2+/Zn2+ sensor proteins, pathological Ca2+/Zn2+ sensors, and Zn2+-resistant Ca2+ sensor proteins.

Serum Calcium Predicts Cognitive Decline and Clinical Progression of Alzheimer's Disease

Ling-Zhi Ma et al.

Summary: This study found a link between serum calcium levels and AD-associated biomarkers, brain structure, cognitive function, and disease progression. Higher serum calcium levels may increase the risk of developing AD in elders, and it can be a useful biomarker in predicting clinical progression in nondemented elders. Further research is needed to explore the underlying mechanisms.

Hydrogen sulfide is neuroprotective in Alzheimer's disease by sulfhydrating GSK3β and inhibiting Tau hyperphosphorylation

Daniel Giovinazzo et al.

Summary: Alzheimer's disease is characterized by memory, executive, and motor function deterioration, with neuropathological hallmarks including neurofibrillary tangles, paired helical filaments, and amyloid plaques. Dysregulation of signaling by hydrogen sulfide has been shown during aging and may play a role in preventing hyperphosphorylation of Tau in Alzheimer's disease. Furthermore, administering the H2S donor sodium GYY4137 to mouse models of Alzheimer's disease has been shown to ameliorate motor and cognitive deficits associated with the disease.

Katarzyna Socha et al.

Summary: The study examined selenium, copper, zinc, and total antioxidant status (TAS) in the serum of patients with Alzheimer's disease (AD) compared to healthy individuals. Lower concentrations of selenium, zinc, and TAS, as well as a higher copper:zinc ratio, were observed in the serum of AD patients. Dietary habits were found to have a significant impact on the concentrations of these parameters in the serum of individuals with AD.

Metallobiology and therapeutic chelation of biometals (copper, zinc and iron) in Alzheimer's disease: Limitations, and current and future perspectives

Kehinde D. Fasae et al.

Summary: Alzheimer's disease is the leading cause of cognitive impairment and dementia globally, with various hypotheses on its pathobiology but no confirmed diagnostics or approved cure. Studies suggest that imbalances in biometal ions and metal-amyloid interactions play crucial roles in AD pathogenesis, highlighting the importance of therapeutic chelation strategies in reducing cerebral metal burden and inhibiting A beta aggregation. Developments in multi-target ligands as potential anti-AD drug candidates show promise in addressing the limitations of current drug candidates and improving chelation-based therapeutics for AD.
