Summary: This study identified targets of OrrA and provided more insights into the role of the orphan response regulator OrrA in Streptomyces through comparing the phenotypes and transcriptomic profiles of wild-type and Delta orrA strains. The interaction of OrrA with the wblA promoter was validated by a pull-down assay, and overexpression of either sco1375 or wblA in Delta orrA partially restored the wild-type phenotype, indicating that these genes mediate some of the effects of OrrA.

A Proteomic Analysis Indicates That Oxidative Stress Is the Common Feature Triggering Antibiotic Production in Streptomyces coelicolor and in the pptA Mutant of Streptomyces lividans

Summary: In most Streptomyces species, antibiotic production is repressed in phosphate proficiency, but the model strain S. coelicolor produces the polyketide antibiotic ACT even more abundantly in phosphate proficiency. The closely related strain S. lividans does not produce ACT in any phosphate condition, but its pptA mutant produces ACT in phosphate limitation. The common feature triggering ACT biosynthesis in S. coelicolor and the pptA mutant of S. lividans is the high oxidative stress.

The involvement of sco1222-23 and sco0921-20 operons in ornithine lipid biosynthesis under phosphate limitation was shown to be positively controlled by the PhoR/PhoP two-component system. Moreover, SCO0920 was found to possess N-acyltransferase activity in addition to O-acyltransferase activity.

Nitric Oxide Signaling for Actinorhodin Production in Streptomyces coelicolor A3(2) via the DevS/R Two-Component System

Summary: Nitric oxide (NO) serves as a signaling molecule in eukaryotic and prokaryotic cells, regulating secondary metabolism in bacteria such as Streptomyces. This study demonstrates the molecular mechanism by which endogenously produced NO controls antibiotic production via the DevS/R two-component system in S. coelicolor A3(2), shedding light on the regulatory mechanisms of NO in bacteria. The findings suggest that studying NO signaling in actinobacteria can aid in the development of strategies against pathogenic actinomycetes and in the actinobacterial industries.

The direct interaction between OhkA and OrrA was revealed, laying a foundation for further elucidation of the molecular mechanism underlying their regulation.

The Response Regulator MacR and its Potential in Improvement of Antibiotic Production in Streptomyces coelicolor

Disruption of the MacRS TCS led to reduced production of ACT, RED, and CDA, and differential gene expression in both secondary and primary metabolism. Moreover, the study indicated a potential role of MacRS in zinc homeostasis and identified multiple MacR binding sites throughout the genome.
