Circulating miRNAs in Type 2 Diabetic Patients with and without Albuminuria in Malaysia

Siti Yazmin Zahari Sham et al.

Summary: This study investigated differentially expressed miRNAs in the sera of T2DM patients in a Malaysian population. The results showed upregulation of miR-874-3p and miR-101-3p in T2DM patients with albuminuria, which correlated with estimated glomerular filtration rate and age. These findings provide evidence for the potential roles of these miRNAs in DKD.

Summary: The study investigated mRNA expression of resistin, TNF-alpha, and miR-124-3p in PBMCs of diabetic patients with and without nephropathy. Results showed increased resistin and TNF-alpha expression in patients with nephropathy, while decreased miR-124-3p expression. These genes may serve as predictive biomarkers for diabetic nephropathy.

Plasma and urinary extracellular vesicle microRNAs and their related pathways in diabetic kidney disease

Sungjin Park et al.

Summary: In this study, we investigated the extracellular vesicle microRNAs (EV miRNAs) and their targets in relation to diabetic kidney disease (DKD). Through small RNA sequencing of plasma and urinary EVs, we identified differentially expressed EV miRNAs and their target mRNAs. These findings were validated in a larger cohort, showing that urinary metabolite concentrations were associated with DKD progression and increased renal expression of NT5E and UGDH proteins in DKD mice models.

Summary: The expression of miR-638 is downregulated in DN and has diagnostic value, negatively correlated with levels of inflammatory factors and positively correlated with urinary albumin excretion and estimated glomerular filtration rate. MiR-638 mimic can inhibit the expression of inflammatory factors and suppress the proliferative ability induced by high glucose.

Urinary sediment microRNAs can be used as potential noninvasive biomarkers for diagnosis, reflecting the severity and prognosis of diabetic nephropathy

Summary: Patients with both diabetes mellitus (DM) and kidney disease could have diabetic nephropathy (DN) or non-diabetic renal disease (NDRD). IgA nephropathy (IgAN) and membranous nephropathy (MN) are the major types of NDRD. Our study identified miR-95-3p, miR-185-5p, miR-1246, and miR-631 as noninvasive biomarkers that can distinguish different patient groups and reflect the severity and prognosis of DN.

Metformin mediates MicroRNA-21 regulated circulating matrix metalloproteinase-9 in diabetic nephropathy: an in-silico and clinical study

Manoj Khokhar et al.

Summary: The study found that levels of hsa-miR-21-5p were upregulated and MMP9 levels were downregulated in T2DM and DN patients. However, in patients treated with metformin, hsa-miR-21-5p was downregulated and MMP9 was upregulated. In-silico analysis revealed the target genes involved in the miR-21 and MMP9 interaction network, suggesting that metformin directly targets miR-21 and regulates MMP9 expression in T2DM patients, influencing the pathogenesis of DN.
