Discovery of BP3 as an efficacious proteolysis targeting chimera (PROTAC) degrader of HSP90 for treating breast cancer

Summary: This study systematically described the design, synthesis, and evaluation of HSP90 degraders based on the PROTAC strategy. The candidate compound 16b (BP3) was found to effectively degrade HSP90 and inhibit the growth of human breast cancer cells. The results suggest that the HSP90-targeting PROTAC strategy has potential novel applications in breast cancer therapy.

DNAJB8 in small extracellular vesicles promotes Oxaliplatin resistance through TP53/MDR1 pathway in colon cancer

Summary: Chemotherapy resistance is a major problem in the treatment of colon cancer. This study found that the heat shock protein 40 family protein DNAJB8 plays a role in L-OHP resistance by inhibiting the degradation of TP53 and promoting MDR1 upregulation. Transfer of DNAJB8 through extracellular vesicles also contributes to colon cancer resistance. Additionally, measuring DNAJB8 levels could serve as a prognostic biomarker and potential therapeutic target for COAD.

Exosomal DNAJB11 promotes the development of pancreatic cancer by modulating the EGFR/MAPK pathway

Summary: This study revealed that exosomal DNAJB11 promotes cell development in pancreatic cancer cells by regulating the expression of HSPA5 and activating the endoplasmic reticulum stress pathway. Additionally, exosomal DNAJB11 can regulate the expression of EGFR and activate the MAPK signaling pathway. Clinical studies suggest that exosomal DNAJB11 has the potential to serve as a diagnostic biomarker and therapeutic target for pancreatic cancer.

DNAJB11 predicts a poor prognosis and is associated with immune infiltration in thyroid carcinoma: a bioinformatics analysis

Summary: The study investigated the prognostic value of co-chaperone protein DNAJB11 in thyroid carcinoma (THCA) using bioinformatics analysis based on TCGA data. It was found that low levels of DNAJB11 mRNA were associated with poor prognosis in THCA, and were significantly correlated with various clinicopathological features. GO enrichment analysis indicated the involvement of DNAJB11 in immune-related biological processes.

DNAJB9 suppresses the metastasis of triple-negative breast cancer by promoting FBXO45-mediated degradation of ZEB1

Summary: The study demonstrates that the restoration of DNAJB9 inhibits migration, invasion, metastasis, and colonization of TNBC cells. Mechanistically, DNAJB9 stabilizes FBXO45 protein and reduces ZEB1 abundance to inhibit EMT and metastasis. Clinical findings show that reduced DNAJB9 expression and FBXO45 abundance in breast cancer tissues are associated with poorer patient outcomes.
