Summary: This study reveals the potential of ginsenosides (GS) in preventing skin photoaging by protecting the skin barrier. The researchers identified the main monomeric constituents of GS and found that GS protects the skin barrier by inhibiting the MAPK signaling pathway, as shown in experiments on hairless mice and HaCaT cells.

Summary: Atopic dermatitis (AD) is a complex skin disease characterized by inflammation and intense itching, with limited effective treatments and potential side effects. Euscaphic acid, derived from natural products, shows anti-inflammatory, anticoagulant, and antioxidant effects in experimental models. It effectively ameliorated AD properties in mouse models by reducing inflammatory cytokines, pruritus factors, and mast cell invasion, suggesting its potential as a therapeutic agent for AD.

Summary: Atopic dermatitis (AD) is a chronic inflammatory skin condition characterized by rashes, itching, and pruritus. The pathogenesis of AD is closely related to immunological factors, including the role of the epidermal barrier and abnormal cytokine expressions. Disrupted epidermal barriers and the abnormal release of pro-inflammatory mediators from keratinocytes, as well as the role of immune cells, contribute to the pathophysiology of AD. Specific biomarkers for diagnosis are lacking, highlighting the need for further research in this area.

Summary: This study found that agerarin reduced IL-4-induced PER2 gene expression in skin cells by suppressing the JAK-STAT3 pathway. Agerarin also restored the rhythmic oscillation of PER2 promoter activity altered by IL-4. These findings suggest that agerarin may have potential therapeutic effects against inflammatory skin conditions associated with disrupted circadian rhythms.

Summary: This article reviews the research progress of marine-derived anticancer drugs, pointing out the rich sources of cytotoxic compounds in marine flora and fauna that have been overlooked. Anticancer drugs derived from marine sources mainly work by stimulating apoptotic signal transduction pathways. The article also provides a comprehensive overview of marine-derived drug development, including sources, modes of action, and discusses future prospects.

Summary: In recent years, the understanding of the pathogenesis of atopic dermatitis (AD) has expanded, leading to the development of novel therapeutic molecules that target the core inflammatory components of the disease. The Janus kinase (JAK)/signal transducer and activation of transcription (STAT) pathway is involved in intracellular signal transduction and gene transcription regulation. The activation of T-helper (Th)-2 and Th22 immune responses plays a pivotal role in the immunopathogenesis of AD, followed by a variable degree of Th1 and Th17 activation. JAK inhibitors have emerged as a potential treatment option for AD due to their ability to inhibit the signal transduction of inflammatory cytokines.

Summary: Atopic dermatitis (AD) is a chronic inflammatory skin condition that greatly impacts the quality of life for affected individuals. Current research is focused on developing topical treatments that target various aspects of AD pathogenesis, such as Janus kinase inhibition, phosphodiesterase-4 inhibition, aryl hydrocarbon modulation, and modulation of the microbiome. Promising preliminary findings on the efficacy and tolerability of these treatments have been reported, but further research is needed.

Summary: This study identified the molecular mechanisms by which inflammatory cytokines inhibit the expression of FLG in the skin. It was found that the formation of the FRA1:c-JUN:HDAC1 complex is promoted under TNFa + IFNγ stimulation, leading to the downregulation of FLG expression. This insight into the regulatory pathways of FLG expression provides potential therapeutic strategies for restoring skin barrier function in chronic skin inflammation.

Summary: The skin serves as a complex immune organ, with keratinocytes playing a crucial role in the pathogenesis of atopic dermatitis. Activation of keratinocytes contributes to immune dysregulation and biological processes that drive the development of atopic dermatitis. Understanding the innate immune functions of keratinocytes can provide insights into the pathophysiological processes of atopic dermatitis and aid in the development of new therapeutic approaches.
