Thermophoretic microfluidic cells for evaluating Soret coefficient of colloidal particles

Namkyu Lee et al.

Summary: Thermodiffusion or thermophoresis has gained significant interest in various engineering fields. This study proposes a thermophoretic microfluidic cell that enables quantitative measurements of the Soret coefficient of colloids. The cell utilizes fluorescence lifetime imaging microscopy to measure the spatial temperature profile and fluorescence intensity to monitor the concentration profile. The measured Soret coefficients with the cell show good agreement with those determined using a Thermal Diffusion Forced Rayleigh Scattering setup, with an average difference of less than 8%.

Binding Thermodynamics and Dissociation Kinetics Analysis Uncover the Key Structural Motifs of Phenoxyphenol Derivatives as the Direct InhA Inhibitors and the Hotspot Residues of InhA

Qianqian Zhang et al.

Summary: In this study, the binding thermodynamic and kinetic information of direct inhibitors of InhA to phenoxyphenol derivatives were explored using multiple computer-aided drug design strategies. The results showed that van der Waals interactions were the main driving force for protein-ligand binding, and key amino acid residues contributed significantly to the binding energy.

Fluorescence Cross-Correlation Spectroscopy Yields True Affinity and Binding Kinetics of Plasmodium Lactate Transport Inhibitors

Iga Jakobowska et al.

Summary: In this study, a novel strategy to combat malaria by blocking lactate export in the parasitic protozoan Plasmodium falciparum was discovered. Small drug-like molecules were identified that inhibit the lactate transporter PfFNT and kill parasites in culture. Biophysical and functional assays were used to determine binding kinetics, affinity, and resistance against mutated PfFNT, providing valuable insights for potential future drug development.

Kinetic Microscale Thermophoresis for Simultaneous Measurement of Binding Affinity and Kinetics

Julian A. C. Stein et al.

Summary: MST is a versatile technique for measuring binding affinities, based on directional movement of molecules in a temperature gradient. We extended MST to measure binding kinetics by increasing thermal dissipation, allowing for simultaneous determination of kinetics and binding affinity. By measuring DNA hybridization rates, we observed linear dependence of on-rates on salt concentration and weak dependence on strand length and temperature.

Structure and mechanism of secondary sulfonamide binding to carbonic anhydrases

Denis Baronas et al.

Summary: Research has shown that secondary sulfonamides bind CA with lower affinity compared to primary sulfonamides, and their binding characteristics exhibit a U-shape dependence similar to primary sulfonamides. Experimental and computational analyses dissected protein binding-linked protonation reactions and atomic contributions to the binding energetics.
