Summary: The ancestor of beta-1,3-xylanases (AncXyl09) was successfully reconstructed using optimized ancestral sequences reconstruction strategy, showing good thermal stability and substrate promiscuity. It can hydrolyze lichen in addition to the original substrate, producing mainly disaccharides which can facilitate the separation and purification of oligosaccharides. This design represents a semi-rationally engineered beta-1,3-xylanase, opening new possibilities for beta-1,3-xylanase engineering, active oligosaccharide preparation, and marine algae resource utilization.
