OtDUB from the Human Pathogen Orientia tsutsugamushi Modulates Host Membrane Trafficking by Multiple Mechanisms

Jason M. Berk et al.

Summary: Bacterial pathogens often manipulate host cell membrane-trafficking pathways for various purposes. In this study, the OtDUB protein from Orientia tsutsugamushi was found to disrupt membrane trafficking through multiple mechanisms, including direct binding to the clathrin adaptor-protein (AP) complexes AP-1 and AP-2, as well as binding to phosphatidylserine. These findings shed light on the role of OtDUB in Orientia infections.

A versatile new tool derived from a bacterial deubiquitylase to detect and purify ubiquitylated substrates and their interacting proteins

Mengwen Zhang et al.

Summary: Protein ubiquitylation is a crucial posttranslational modification, and the OtUBD tool offers an efficient way to detect and purify ubiquitylated proteins. It enables selective purification of covalently ubiquitylated proteins or co-isolation of both ubiquitylated proteins and their interacting proteins.

Functional assignment to hypothetical proteins in Orientia tsutsugamushi strain Ikeda

Dixit Sharma et al.

Summary: Orientia tsutsugamushi is an intracellular bacterial pathogen that causes zoonotic scrub typhus in humans. The genome of O. Tsutsugamushi contains 214 hypothetical proteins, of which the functions of 44 have been annotated. These annotated proteins are classified into five main classes and their computational analysis provides insights into potential therapeutic targets for understanding their roles in various biological and cellular processes, including pathogenesis.

Cells within cells: Rickettsiales and the obligate intracellular bacterial lifestyle

Jeanne Salje

Summary: This paragraph provides an overview of the Rickettsiales group of obligate intracellular vector-borne Gram-negative bacteria, discussing their biology and interactions with host cells. It focuses on pathogenic species or those important for human health, detailing genomic information, bacterial cell biology, intracellular lifestyles, and immune response evasion mechanisms. Salje describes key features of pathogenic Rickettsiales species and their molecular interactions with mammalian host cells.

Orientia tsutsugamushi Nucleomodulin Ank13 Exploits the RaDAR Nuclear Import Pathway To Modulate Host Cell Transcription

Haley E. Adcox et al.

Summary: Orientia tsutsugamushi is an intracellular pathogen causing scrub typhus, with Ank13 being a key effector that enters the nucleus using RaDAR nuclear import mechanism. Ank13 dysregulates the expression of over 2,000 host genes, particularly those involved in transcriptional control and the inflammatory response, through a combination of nucleotropism and F-box-dependent mechanisms. This effector plays a crucial role in the pathogenesis of scrub typhus by reprogramming host cell transcription.
