Summary: Patients with psoriasis, especially psoriatic arthritis, have an increased risk of developing osteoporosis due to shared risk factors like vitamin D deficiency and chronic inflammation. The IL-33/ST2 axis, along with vitamin D, plays a key role in bone remodeling and skin barrier pathophysiology.IL-33 and vitamin D stimulate the production of anti-osteoclastogenic cytokines, influencing regulatory and Th2 cells to prevent osteoclastogenesis.

Summary: This study investigated the effects of vitamin D deficiency on the expression of inflammatory mediators in cartilage, finding that deficiency is associated with increased expression while supplementation decreases expression. It was also discovered that vitamin D deficiency leads to an increase in M1 macrophages, while supplemented cartilage showed an increase in M2 macrophages. Additionally, calcitriol was found to promote M2 macrophage polarization. Overall, modulating the expression of inflammatory mediators with vitamin D supplementation may serve as a novel therapeutic approach to reduce inflammation and cartilage degeneration in osteoarthritis.
