Comprehensive Genomic Profiling of Neuroendocrine Carcinomas of the Gastrointestinal System

Shinichi Yachida et al.

Summary: Gastrointestinal neuroendocrine carcinoma (GIS-NEC) is a rare but highly malignant tumor that is genetically distinct from gastrointestinal neuroendocrine tumors (GIS-NET). There are clear genomic differences between pancreatic NECs (Pant-NEC) and nonpancreatic GIS-NECs (Nonpanc-NEC). Panc-NECs can be classified into two subgroups based on genomic and epigenomic features.

Non-functional pancreatic neuroendocrine tumours: ATRX/DAXX and alternative lengthening of telomeres (ALT) are prognostically independent from ARX/PDX1 expression and tumour size

Summary: Recent studies have shown that ATRX/DAXX and ALT are important prognostic factors for NF-PanNETs, especially in tumors less than or equal to 2.0 cm. These biomarkers are highly specific for pancreatic origin among NET metastases of unknown primary.

ALT Positivity in Human Cancers: Prevalence and Clinical Insights

Danny MacKenzie et al.

Summary: The Alternative Lengthening of Telomeres (ALT) pathway has been recognized for its clinical significance in cancer development, diagnosis, and treatment. This review examines the activation of ALT, its prevalence in different cancer types, and its impact on prognosis and pathogenetics. Additionally, advancements in understanding ALT at the molecular level and potential therapies targeting ALT are highlighted.

Alternative Lengthening of Telomeres (ALT) in Pancreatic Neuroendocrine Tumors: Ready for Prime-Time in Clinical Practice?

Claudio Luchini et al.

Summary: In pancreatic neuroendocrine tumors (PanNET), ALT is strongly correlated with the mutational status of two chromatin remodeling genes, DAXX and ATRX. ALT emerged as a reliable indicator of worse prognosis for PanNET, aiding in clinical stratification and identification of high-risk patients, and supporting the pancreatic origin of neuroendocrine tumors for improving diagnostic workflow in patients with neuroendocrine metastasis from unknown primary.
