A proof-of-principle study on implementing polymerase chain displacement reaction (PCDR) to improve forensic low-template DNA analysis

Yuguo Huang et al.

Summary: PCR plays a crucial role in forensic DNA analysis, but often faces challenges with low-template DNA samples. Polymerase chain displacement reaction (PCDR) is an efficient technique combining PCR and strand displacement reaction. This study demonstrated that PCDR can improve the efficiency and multiplexing potential in forensic LTDNA analysis.

MPSproto: An extension of EuroForMix to evaluate MPS-STR mixtures

Oyvind Bleka et al.

Summary: The study found that the MPSproto models performed better in handling stutter artefacts and other typing errors commonly found in MPS-STR data compared to the traditional EuroForMix model. The gamma model implemented in MPSproto showed the best performance and was adequate for sequence reads when a low analytical threshold was applied.

STRNaming: Generating simple, informative names for sequenced STR alleles in a standardised and automated manner

Jerry Hoogenboom et al.

Summary: The STRNaming algorithm generates human-readable names for sequenced STR alleles by using a reference sequence at each locus and automatically assigning a unique, sequence-descriptive name that also includes the capillary electrophoresis allele number. Settings for STRNaming were established based on international preferences in the forensic community, ensuring that small changes in the sequence correspond to small changes in the allele name, aiding in recognizing stutter products and other sequence variants.
