Summary: The study reveals that excessive growth hormone (GH) induces TGF-beta 1-dependent Notch1 signaling leading to mitotic catastrophe in podocytes. Inhibition of JAK2, TGFBR1, or Notch can prevent podocytes from reentering the cell cycle and protect them from cell death. The aberrant GH signaling in podocytopathy highlights the potential therapeutic application of TGF-beta 1 or Notch inhibitors for diabetic nephropathy (DN).
