Reimagining cholinergic therapy for Alzheimer's disease

Ezio Giacobini et al.

Summary: The review discusses the use of cholinesterase inhibitors in the treatment of Alzheimer's disease, the role of the basal forebrain cholinergic system in cognition, and potential future therapies targeting the NGF metabolic pathway and M1 muscarinic receptor. It provides an updated view of the cholinergic-oriented therapy of Alzheimer's disease and suggests new alternatives to enhance its clinical effect.

Advancing combination treatment with cilostazol and caffeine for Alzheimer's disease in high fat-high fructose-STZ induced model of amnesia

Adel A. Gomaa et al.

Summary: Caffeine and combined treatment of caffeine and CSZ can effectively improve cognitive impairment related to high fat/high fructose fed T2D rats and protect neurons from damage. Additionally, caffeine and CSZ combined treatment can normalize the accumulation of Aβ and phosphorylated tau protein in the brain, regulate glutamate gene expression, cholinesterase level, caspase-3 activity, and levels of oxidative stress and pro-inflammatory cytokines. Caffeine enhances the inhibitory effects of CSZ on insulin resistance, GSK-313 activity, hypercholesterolemia, oxidative stress, and pro-inflammatory cytokines.

miRNA-338-3p/CAMK IIα signaling pathway prevents acetaminophen-induced acute liver inflammation in vivo

Chen Zhang et al.

Summary: This study demonstrated that miR-338-3p plays a crucial role in APAP-induced acute liver injury by inhibiting inflammation signaling pathways, thus alleviating liver injury caused by APAP overdose. Additionally, it was found that calcium/calmodulin-dependent protein kinase II alpha (CAMK II alpha) is a direct target of miR-338-3p.

Protective effect of ginsenoside Rh2 on scopolamine-induced memory deficits through regulation of cholinergic transmission, oxidative stress and theERK-CREB-BDNFsignaling pathway

Jingwei Lv et al.

Summary: The study demonstrated that Rh2 treatment effectively improved memory deficits induced by Scop in mice by modulating the cholinergic system, inhibiting oxidative stress, and activating the ERK-CREB-BDNF signaling pathway. These findings suggest that Rh2 may be a promising candidate compound for treating Alzheimer's disease.

Potential Repositioning of Anti-cancer EGFR Inhibitors in Alzheimer's Disease: Current Perspectives and Challenging Prospects

Heba M. Mansour et al.

Summary: Recent studies have shown that EGFR inhibition may have neuroprotective effects on Alzheimer's disease (AD), but the lack of exploration on signaling transductions, poor blood-brain barrier permeability of tested drugs, and disappointing clinical trial outcomes have hindered conclusive evidence of the efficacy of EGFR inhibitors in AD treatment. This has raised debate on the potential involvement of EGFR inhibitors in future clinical trials.
