Sustainable and efficient skin absorption behaviour of transdermal drug: The effect of the release kinetics of permeation enhancer

Summary: This study investigated how the release kinetics of permeation enhancers affected their enhancement efficacy on drug skin absorption. It was found that the release of acidic drugs followed the Higuchi release model with short duration, resulting in unsustainable enhancement efficiency. On the other hand, the release of basic drugs followed a zero-order model with long duration, leading to sustainable and efficient enhancement. Understanding the release kinetics of enhancers can help in the selection and optimization of transdermal patches.

A Systematic Quantitative Evaluation of Permeation Enhancement Window: Transdermal Permeation Enhancing Dynamics Establishment and Molecular Mechanisms Characterization of Permeation Enhancer

Summary: Currently, there is limited research on the dynamics of transdermal permeation enhancement of permeation enhancers. In this study, the dynamics were established based on the content of enhancer Plurol Oleique CC in the skin and the increment of drug permeation amount. A new concept called permeation enhancement window was used to evaluate the enhancement effect of POCC for different drugs. The study found that the differences in the permeation enhancement window were due to the varied interaction strengths among POCC, drugs, and adhesives, as well as changeable SC lipid fluidity.

The effect of pH and salt on the molecular structure and dynamics of the skin

The highest molecular mobility in both lipid and protein components is observed at the lowest pH of 4.0. The study results provide new insights on how SC molecular properties respond to solution conditions, which can be beneficial in developing topical drug delivery and cosmetic formulations.

Comparison of Synthetic Membranes to Heat-Separated Human Epidermis in Skin Permeation Studies In Vitro

Summary: This study compared permeation across innovative synthetic membranes and human epidermis, with the Strat-M membrane showing the greatest similarity to HSE. Good correlations were found between the permeation data of different formulations across different membranes, allowing the prediction of permeation across HSE using synthetic membranes. Strat-M and Skin PAMPA membranes have the potential to select and differentiate dermal formulations containing diclofenac sodium as an early screening model.

An investigation on the effect of drug physicochemical properties on the enhancement strength of enhancer: The role of drug-skin-enhancer interactions

Different drugs showed varied enhancement ratios, with drugs with lower polar surface areas and polarizability exhibiting more pronounced enhancements when interacting with the enhancer POCC in transdermal applications.
