Nontraditional Fragment Couplings of Alcohols and Carboxylic Acids: C(sp3)-C(sp3) Cross-Coupling via Radical Sorting

Holt A. Sakai et al.

Summary: Alcohols and carboxylic acids are important functional groups in organic chemistry that can undergo radical activation and transition metal catalysis under visible light photoredox catalysis. In this report, a dual combination of NHC-mediated deoxygenation and hypervalent iodine-mediated decarboxylation is used to achieve the C(sp3)-C(sp3) cross-coupling of alcohols and carboxylic acids. This method offers a mild and practical approach for the synthesis of alkyl-alkyl cross-coupled products.

FeIII/TBHP mediated remote C-O bond construction of 8-aminoquinolines: access to methoxylation and cyanomethoxylation

Mengfei Zhao et al.

Summary: Novel approaches for C5-selective methoxylation and cyanomethoxylation of 8-aminoquinolines under the Fe-III/TBHP system were developed. Various substituted quinolines were tolerated to yield the corresponding ether products. A plausible mechanism involving the addition of alkoxyl radicals to an iron-chelated intermediate was proposed.

Remote selective decarboxylative difluoroarylmethylation of 8-aminoquinolines under transition metal-free conditions

Xiaolong Guo et al.

Summary: A facile transition metal-free decarboxylative C4 selective C-H difluoroarylmethylation of 8-aminoquinolines under simple aqueous conditions has been developed, providing a novel and concise route for the construction of a difluoromethylene linker between (hetero)aromatic rings (Ar-CF2-Ar). Initial mechanistic studies suggest the involvement of a reasonable radical coupling reaction in this C4 selective C-H difluoroarylmethylation.

Iron-Catalyzed C-C Single-Bond Cleavage of Alcohols

Wei Liu et al.

Summary: An iron-catalyzed deconstruction/hydrogenation reaction of alcohols through C-C bond cleavage has been developed via photocatalysis, yielding ketones or aldehydes as products. This process is applicable to tertiary, secondary, and primary alcohols with a wide range of substituents, including complex natural alcohols. Investigating the mechanism uncovered a process involving chlorine radical-enhanced O-H homolysis, assisted by 2,4,6-collidine.

Iron-Catalyzed Photoinduced Remote C(sp3)-H Amination of Free Alcohols

Ni Xiong et al.

Summary: The experiment presents a general photocatalytic method for the remote C(sp(3))-H bond amination of free aliphatic alcohols, utilizing electron transfer between FeCl3 catalyst and alkanols under blue LED irradiation. It enables alkoxy radical formation under mild redox-neutral conditions without the need for additional oxidants and prefunctionalization. The subsequent selective 1,5-hydrogen atom transfer (HAT) and amination offer a simple and efficient way to access molecular complexity from readily available and bulk alcohols.

Copper Catalyzed C(sp3)- H Bond Alkylation via Photoinduced Ligand-to-Metal Charge Transfer

Sean M. Treacy et al.

Summary: By utilizing catalytic CuCl2, various feedstock chemicals can be functionalized through the coupling of unactivated C(sp(3))-H bonds with electron-deficient olefins. This reaction, enabled by Ligand-to-Metal Charge Transfer (LMCT), generates a chlorine radical capable of abstracting strong electron-rich C(sp(3))-H bonds. The protocol offers a simple operation and atom economical approach to generate a highly active hydrogen atom transfer reagent.

Metal-Free α-C(sp3)-H Aroylation of Amines via a Photoredox Catalytic Radical-Radical Cross-Coupling Process

Guo-Qiang Xu et al.

Summary: The metal-free C(sp(3))-H aroylation of amines via visible-light photoredox catalysis provides a direct route for constructing a useful a-amino aryl ketone skeleton. Selected products show good biological activity for protecting PC12 cells, indicating the potential of this skeleton as a new neuroprotective agent. Mechanism experiments suggest that this transformation follows a photoredox catalytic radical-radical cross-coupling pathway.

Selectfluor-Enabled C(sp3)-H Alkoxylation of 3-Methylfuranocoumarins

Zengxin Qin et al.

Summary: A metal-free method for the direct C(sp(3))-H bond alkoxylation of 3-methylfuranocoumarins with alcohols has been developed. Selectfluor was used to enable the (hetero)benzylic C-H etherification by tuning the reaction conditions. Various alcohols were found to be compatible in this transformation, yielding suitable products. Mechanistic studies suggest that the reaction may involve a double addition process of alcohols, as well as the departure of a fluoride anion and the formation of an oxonium ion.

Metallaphotoredox-enabled deoxygenative arylation of alcohols

Zhe Dong et al.

Summary: Metal-catalysed cross-couplings are widely used for the formation of C-C bonds, but alkyl cross-couplings using alcohols as coupling partners remain relatively underdeveloped. A new method for direct deoxygenative cross-coupling of free alcohols has been developed, showing potential for a wide range of applications, including the pharmaceutical industry. This technology represents a general strategy for combining in situ alcohol activation with transition metal catalysis.

Jianyang Dong et al.

Summary: A mild and operationally simple method using vinyl ethers as acetyl sources for Minisci C-H acetylation of N-heteroarenes is reported. This method does not require conventional photocatalysis, electrocatalysis, metal catalysis, light activation, or high temperature, making it more sustainable in terms of cost, reagent toxicity, and waste generation compared to previous methods. It is expected to obtain medically relevant molecules from abundant feedstock materials.

Photocatalytic C-H activation and the subtle role of chlorine radical complexation in reactivity

Qiaomu Yang et al.

Summary: The reported carbon-hydrogen (C-H) activation of alkanes is actually mediated by the photocatalyst [NEt4](2)[CeCl6], and RO center dot are not intermediates. The generation of chlorine radical (Cl center dot) in the photocatalytic system is found to be the rate-limiting step. Density functional theory calculations support the formation of [Cl center dot][alcohol] adducts, which can manifest a masked RO center dot character.

A cross-dehydrogenative C(sp3)-H heteroarylation via photo-induced catalytic chlorine radical generation

Chia-Yu Huang et al.

Summary: Hydrogen atom abstraction from C(sp(3))-H bonds of naturally abundant alkanes for alkyl radical generation represents a promising yet underexplored strategy in the alkylation reaction designs. The authors demonstrate a photo-induced and chemical oxidant-free cross-dehydrogenative coupling between alkanes and heteroarenes using catalytic chloride and cobalt catalyst, achieving successful couplings with satisfactory yields. The dual catalytic platform features the in situ engendered chlorine radical for alkyl radical generation and exploits the cobaloxime catalyst for catalytic turnover enabling hydrogen evolution.
