Angiogenesis and anti-leukaemia activity of novel indole derivatives as potent colchicine binding site inhibitors

Summary: This article reports the effects of the screened DYT-1 compound from an in-house library on inhibiting tubulin polymerisation, anti-angiogenesis, and anti-leukaemia proliferation in vitro and in vivo. Further investigation led to the discovery of compound 29e, which displayed excellent activity in tubulin and zebrafish assays, and nanomolar potency against a variety of leukaemia cell lines. Compound 29e stably binds to the tubulin colchicine site, showing promising anti-angiogenesis and anti-leukaemia effects. These results suggest that 29e could be considered as an effective anti-angiogenesis and anti-leukaemia drug candidate.

Summary: A library of novel 1,3,4-oxadiazole bioisosteres was synthesized and evaluated for their cytotoxic activity. Several of the new compounds showed potent anticancer activity, surpassing the previously synthesized oxadiazole compound. The nature of the selenadiazole and thiadiazole rings may play a crucial role in the antitumor activity. These compounds exhibited strong cytotoxic effects in tumor cells compared to human primary cells.

Design, semi-synthesis and bioactivity evaluation of novel podophyllotoxin derivatives as potent anti-tumor agents

Summary: A series of potential candidate molecules with excellent antitumor activity targeting tubulin and PTEN/PI3K/Akt signaling pathway were synthesized by modifying the molecule structure of podophyllotoxin (PPT) at the C-4 position. Compound 12c showed stronger antiproliferative activities against HGC-27, MCF-7, and H460 cell lines than etoposide (VP-16), with lower toxicity in healthy human cells. Further analysis revealed that 12c disrupted the dynamic equilibrium of microtubules and inhibited the process of epithelial-mesenchymal transition. The downregulation of PTEN/PI3K/Akt signaling pathway and cascade influence on the mitochondrial pathway contributed to the apoptosis of cancer cells. Therefore, 12c has the potential to be a candidate molecule for gastric cancer clinical treatment.

Discovery of new indole-based 1,2,4-triazole derivatives as potent tubulin polymerization inhibitors with anticancer activity

Summary: A series of novel indole-based 1,2,4-triazole derivatives were designed and synthesized, with compound 9p showing excellent antiproliferative efficacy against HeLa cells in the nanomolar range and no cytotoxic activity towards normal cells. Mechanism analysis revealed that 9p arrested the cell cycle and induced apoptosis in HeLa cells, while also effectively inhibiting tubulin polymerization. The study suggests that indole-triazole hybrids could serve as a promising scaffold for developing novel tubulin polymerization inhibitors for cancer treatment.
