Targeting the Central Pocket of the Pseudomonas aeruginosa Lectin LecA

Investigation of Direct and Retro Chromone-2-Carboxamides Based Analogs of Pseudomonas aeruginosa Quorum Sensing Signal as New Anti-Biofilm Agents

Summary: Biofilm formation is a major cause of therapeutic failure and poses significant challenges to public health and the economy. Pseudomonas aeruginosa, in particular, is responsible for severe infections due to its ability to form resistant biofilms. In this study, chromone-based compounds were evaluated as potential ligands for PqsR, a receptor involved in biofilm formation, and a library of chromone carboxamides was synthesized and tested for their ability to inhibit biofilm formation. Several compounds showed promising results as inhibitors of P. aeruginosa biofilms, highlighting the potential for new therapeutic approaches.

Lectin-Targeted Prodrugs Activated by Pseudomonas aeruginosa for Self-Destructive Antibiotic Release

Summary: In this study, lectin-targeted antibiotic prodrugs against Pseudomonas aeruginosa were synthesized by conjugating several fluoroquinolones to lectin probes. These prodrugs remained non-toxic in systemic distribution and were activated to exert bactericidal effects only at the infection site. Additionally, the prodrugs exhibited good ADME properties and reduced toxicity.

Non-Carbohydrate Glycomimetics as Inhibitors of Calcium(II)-Binding Lectins

Summary: Due to the antimicrobial resistance crisis, lectins are considered novel drug targets, with the study focusing on non-carbohydrate inhibitors for LecA. A catechol was identified as an experimentally confirmed hit, acting as a millimolar LecA inhibitor. The first co-crystal structure of a non-carbohydrate inhibitor in complex with a bacterial lectin demonstrated how the catechol mimics the binding of natural glycosides with LecA, indicating a fundamentally new class of glycomimetics.

Summary: In this study, a series of novel rotaxanes were synthesized, showing potent inhibition of biofilm growth in Pseudomonas aeruginosa. The synergistic action of the three subunits in the glycorotaxanes was demonstrated, with the best compound's antibiofilm activity being lectin-mediated, and none of the final rotaxanes were bactericidal.
