Pharmaceutical pollution of the world's rivers

John L. Wilkinson et al.

Summary: Environmental exposure to active pharmaceutical ingredients (APIs) can have negative effects on ecosystems and human health. A global-scale study of API pollution in rivers highlights the highest concentrations in sub-Saharan Africa, south Asia, and South America, primarily in low- to middle-income countries with poor wastewater and waste management infrastructure. The most frequently detected APIs include carbamazepine, metformin, and caffeine, with some concentrations exceeding safe levels for aquatic organisms. Pharmaceutical pollution poses a global threat to environmental and human health and has implications for achieving the United Nations Sustainable Development Goals.

Chronic Embryo-Larval Exposure of Fathead Minnows to the Pharmaceutical Drug Metformin: Survival, Growth, and Microbiome Responses

Joanne L. Parrott et al.

Summary: The study found that metformin at environmentally relevant concentrations (3.44 and 33.6 μg/L) and at 10 times municipal wastewater effluents concentrations (269 μg/L) did not have adverse effects on larval growth or gut microbiome in this common freshwater fish species.

Neuroendocrine pathways at risk? Simvastatin induces inter and transgenerational disruption in the keystone amphipod Gammarus locusta

T. Neuparth et al.

Summary: The primary focus of environmental toxicological studies is often on the direct effects of chemicals on exposed organisms, while the effects on subsequent non-exposed generations are often overlooked. In this study, the effects of simvastatin on a keystone crustacean were investigated, and it was found that simvastatin at environmentally relevant concentrations has significant inter and transgenerational effects on the neuroendocrine regulation of crustaceans, leading to adverse effects on reproduction and growth.

Low Levels of Perfluorooctanoic Acid Exposure Activates SteroidHormone Biosynthesis through Repressing Histone Methylation in Rats

Xuejingping Han et al.

Summary: This study investigated the effects of low-dose PFOA exposure on testicular steroidogenesis in rats and revealed the role of histone modifications. The results showed that PFOA exposure increased the levels of certain steroid hormones and up-regulated the expression of the key gene Star, while down-regulating other steroidogenic genes. Additionally, PFOA exposure significantly reduced multiple histone modifications, suggesting an epigenetic mechanism for PFOA's disruption of male reproductive endocrine.

Comparative Effects of Embryonic Metformin Exposure on Wild and Laboratory-Spawned Fathead Minnow (Pimephales promelas) Populations

Kristin M. Nielsen et al.

Summary: Metformin, commonly used for treating Type 2 diabetes, is frequently detected in aquatic ecosystems. Laboratory studies have shown that exposure to environmental concentrations of metformin can affect metabolic pathways and impact the growth of early life stage fish. It is unknown if these effects occur in wild populations. This study evaluates the representativeness of laboratory studies and compares the sensitivities of wild and lab-spawned fish. The results indicate that developmental metformin exposure affects the growth and morphology of both cohorts, with wild-spawned fish being more affected. The study also confirms the bioavailability of metformin in natural systems and suggests that current exposure scenarios may negatively impact developing fish.

Effects of Metformin and its Metabolite Guanylurea on Fathead Minnow (Pimephales promelas) Reproduction

Brett R. Blackwell et al.

Summary: Multiple experiments were conducted to investigate the reproductive toxicity of metformin and guanylurea to adult fathead minnows. The results showed that both compounds did not have significant effects on steroidogenesis, gene transcription, hormone concentrations, or fecundity. The impacts on the hepatic transcriptome and metabolome were time-dependent, with the greatest effect observed after 23 days of exposure to 100 μg/L guanylurea.
