Importance of Rare DPYD Genetic Polymorphisms for 5-Fluorouracil Therapy in the Japanese Population

Summary: In this study, an in vitro analysis was conducted on 41 DPD allelic variants to investigate changes in enzymatic activity, with 7 variants showing significantly decreased activity and 2 variants displaying no enzymatic activity. Our findings suggest that DPD dimerization is essential for enzymatic activity and these variants may contribute to the observed inter-individual variability in the pharmacokinetics and pharmacodynamics of 5-FU. Additionally, rare DPYD variants, although at low frequencies, could serve as important pharmacogenomic markers associated with severe 5-FU toxicity in the Japanese population.

Study Profile of the Tohoku Medical Megabank Community-Based Cohort Study

Summary: The study established a community-based cohort to assess the long-term impact of the Great East Japan Earthquake on disaster victims and gene-environment interactions on major disease incidence. Results showed that 70% of participants joined at health check-up sites, Type 1 survey participants were more likely to have psychological distress, women were more likely to have psychological distress than men, and coastal residents had higher levels of psychological distress compared to inland residents.

jMorp updates in 2020: large enhancement of multi-omics data resources on the general Japanese population

Summary: The Tohoku Medical Megabank project conducted genome and omics analyses on participants from two cohort studies, resulting in the creation of jMorp, a web-based database providing genome, methylome, transcriptome, and metabolome data. This resource, which includes Japanese reference sequences and allele frequencies, as well as concentration distributions of over 755 metabolites, will aid in the discovery of disease biomarkers and personalized disease prevention and early diagnosis in the general population.
