Synthesis and evaluation of a novel adapter lipid derivative for preparation of cyclic peptide-modified PEGylated liposomes: Application of cyclic RGD peptide

Naoya Kato et al.

Summary: Peptide ligand modified nanoparticles can be easily prepared using the post-insertion method, which requires highly water dispersible peptide-lipid conjugates. In this study, a novel adapter lipid derivative was synthesized using the click reaction, enabling the synthesis of various cyclic peptides and the preparation of cRGD-modified PEGylated liposomes with good water dispersibility. The resulting liposomes efficiently associated with integrin αvβ3-expressing colon carcinoma cells and exhibited high cytotoxicity when loaded with doxorubicin.

Synthesis and pharmacokinetic characterisation of a fluorine-18 labelled brain shuttle peptide fusion dimeric affibody

Takahiro Morito et al.

Summary: Brain PET imaging using radiolabelled proteins is a promising approach for visualizing molecular targets in the brain, but faces challenges in pharmacokinetics and labelling methods. This study demonstrates the successful synthesis of a BBB-permeable affibody molecule radiolabelled with a unique fluorine-18 labelling method. The results suggest that small brain shuttle peptide-fused fluorine-18 labelled protein binders may have potential applications in brain molecular imaging.

Rethinking CRITID Procedure of Brain Targeting Drug Delivery: Circulation, Blood Brain Barrier Recognition, Intracellular Transport, Diseased Cell Targeting, Internalization, and Drug Release

Shaobo Ruan et al.

Summary: This review analyzes the typical delivery process of nanoparticle-based brain-targeting drug delivery systems, proposing a six-step CRITID delivery cascade to provide a deep understanding of the issues and requirements for designing efficient and specific systems. Strategies for troubleshooting these issues are reviewed and state-of-the-art design features are highlighted to guide the development of more effective brain-targeting drug delivery systems.

B3Pdb: an archive of blood-brain barrier-penetrating peptides

Vinod Kumar et al.

Summary: The blood-brain barrier poses challenges in treating brain-related ailments. Peptides-based therapeutics have shown promise, but a comprehensive knowledge base on their use as therapeutic molecules for brain-related disorders is lacking. The B3Pdb database provides essential information on BBB-penetrating peptides, facilitating the rational design of therapeutics for CNS-related disorders.

Temozolomide nano enabled medicine: promises made by the nanocarriers in glioblastoma therapy

P. Soma Yasaswi et al.

Summary: GBM, a life-threatening IV grade glioma brain cancer, remains a challenge to treat due to the toxic nature and fast degradation of TMZ, along with lack of targeting ability increasing chances of systemic toxicities. Nano-enabled technology improves the efficiency of TMZ and reduces toxicity to healthy tissues.

Post-capillary venules are the key locus for transcytosis-mediated brain delivery of therapeutic nanoparticles

Krzysztof Kucharz et al.

Summary: Limited understanding of interactions between nanoparticle drug carriers and the blood-brain barrier underlies many translational failures in treatments of brain disorders. Here the authors use two-photon microscopy in mice to characterize the receptor-mediated transcytosis of nanoparticles at all steps of delivery from the blood to the brain in vivo.
