Comparison of dystrophin expression following gene editing and gene replacement in an aged preclinical DMD animal model

Niclas E. Bengtsson et al.

Summary: This study explores the role of gene editing in an older DMD animal model and compares it to gene replacement. The study found that gene editing could restore the dystrophin reading frame in a portion of the genome, but gene replacement had better effects. This suggests that the efficacy of CRISPR-based therapies is linked to the pathological state at the time of treatment.

TRAPPC11-related muscular dystrophy with hypoglycosylation of alpha-dystroglycan in skeletal muscle and brain

Pinki Munot et al.

Summary: This study presents five cases of early-onset TRAPPC11-related muscular dystrophy, with muscle pathology findings in all cases, post-mortem brain pathology findings in one case, and membrane trafficking assays in another. The findings suggest that recessive mutations in TRAPPC11 are linked to muscular dystrophies with hypoglycosylation of alpha-dystroglycan, and structural cerebellar involvement similar to that seen in N-linked congenital disorders of glycosylation.

Exocyst-mediated membrane trafficking of the lissencephaly-associated ECM receptor dystroglycan is required for proper brain compartmentalization

Andriy S. Yatsenko et al.

Summary: Studies have shown that dystroglycan has a spatiotemporally dynamic expression pattern in the brain during development, and proteins in the exocyst complex are associated with Dg in neurons. These findings help further understand the regulatory mechanisms of Dg in the brain.

β-Dystroglycan Restoration and Pathology Progression in the Dystrophic mdx Mouse: Outcome and Implication of a Clinically Oriented Study with a Novel Oral Dasatinib Formulation

Paola Mantuano et al.

Summary: The study showed that a new oral DAS/HP-beta-CD complex efficiently distributed in mdx mice plasma and tissues in a dose-related fashion, reducing beta-DG phosphorylation and restoring its levels dose-dependently in both diaphragm and gastrocnemius muscle. However, although there was modest improvement in in vivo neuromuscular function, ex vivo muscle force, and histopathology, there was partial recovery of muscle elasticity and a decrease of CK and LDH plasma levels, suggesting an increased sarcolemmal stability of dystrophic muscles. Further research is needed to understand the limitations of DAS effectiveness in dystrophic settings and its potential synergy with dystrophin-based molecular therapies.

Genomic knockout of hsp23 both decreases and increases fitness under opposing thermal extremes in Drosophila melanogaster

Xinyue Gu et al.

Summary: Under exposure to harmful environmental stresses, organisms exhibit a general stress response involving upregulation of heat shock proteins. This study explored the pleiotropic effects of the hsp23 gene in Drosophila melanogaster using the CRISPR/Cas9 system, showing that knockout of hsp23 gene can impact fitness and tolerance in flies.

The burden of heat-related mortality attributable to recent human-induced climate change

A. M. Vicedo-Cabrera et al.

Summary: Current and future climate change is projected to have significant impacts on human health through increasing temperatures. A study found that approximately 37% of warm-season heat-related deaths can be attributed to anthropogenic climate change. Urgent mitigation and adaptation strategies are needed to minimize the public health impacts of climate change.

High degree of conservation of the enzymes synthesizing the laminin-binding glycoepitope of α-dystroglycan

Maria Giulia Bigotti et al.

Summary: The DG complex plays a key role in muscle stability in Metazoa, with mutations in enzymes involved in glycosylation of alpha-DG leading to severe neuromuscular disorders. Investigating the evolution of these modifying enzymes reveals the importance of glycosylation in modulating the connection between sarcolemma and basement membranes, ultimately supporting skeletal muscle stability and movement.
