Phase separation of RNA-binding protein promotes polymerase binding and transcription

Wen Shao et al.

Summary: A new study suggests that RNA-binding proteins constitute half of the chromatin proteome in embryonic stem cells, with some colocalizing with RNA polymerase II at promoters and enhancers. These RNA-binding proteins inhibit premature release of Pol II and enhance the formation of transcription condensates using RNA as multivalent molecules, ultimately promoting polymerase engagement and activity. The study proposes a synergistic interplay between promoter-associated RNAs and their binding proteins to facilitate active transcription through phase separation of polymerase condensates.

RNA-Mediated Feedback Control of Transcriptional Condensates

Jonathan E. Henninger et al.

Summary: This study discusses the mechanisms of feedback control in regulating biological processes, especially in transcription regulation. Research indicates that RNA in transcription initiation promotes condensate formation, while RNA produced during elongation stimulates condensate dissolution. Therefore, transcriptional regulation involves a feedback mechanism where transcribed RNA both stimulates and arrests the process.

Spatial organization of transcribing loci early activation in Drosophila

Shao-Kuei Huang et al.

Summary: Studying early Drosophila embryos revealed that RNA polymerase II accumulates at histone locus bodies during early genome activation. Co-regulated genes do not cluster into nuclear sub-domains, but transcribing genes in close 3D proximity have distinct RNA polymerase II speckles. This challenges the emerging model of clustered active genes in gene regulation.

Co-condensation between transcription factor and coactivator p300 modulates transcriptional bursting kinetics

Liang Ma et al.

Summary: The coactivator p300 can dynamically coordinate its functions by forming clusters with transcription factors, enabling spatially defined activation of its catalytic activity. This co-condensation mechanism modulates the transcriptional initiation rate and burst duration of target genes, contributing to nonlinear gene regulation. The results suggest a mechanism where TF and p300 co-condensation contributes to transcriptional bursting regulation and cooperative gene control.

Transcription organizes euchromatin via microphase separation

Lennart Hilbert et al.

Summary: This study observed the dynamics of euchromatin organization in pluripotent zebrafish cells during transcription initiation, revealing that accumulation of RNA induces the formation of transcriptionally active regions that tend to separate from transcriptionally inactive euchromatin, forming smaller scale microphases.
