Lipid mediator n-3 docosapentaenoic acid-derived protectin D1 enhances synaptic inhibition of hippocampal principal neurons by interaction with a G-protein-coupled receptor

Apostolos Mikroulis et al.

Summary: Epilepsy is a severe neurological disease that affects about 1% of the population. Current drug treatments are ineffective for up to 40% of patients, highlighting the need for novel therapies. A molecule called PD1(n-3DPA) has been found to reduce seizures, cell loss, and cognitive deficits in a mouse model of epilepsy. Additionally, PD1(n-3DPA) may directly affect neuronal activity by modulating synaptic inputs.

Dietary alpha-linolenic acid reduces platelet activation and collagen-mediated cell adhesion in sickle cell disease mice

Simona Stivala et al.

Summary: In summary, the study found that a high-ALA diet could reduce the number of sickle cells in blood smear, liver fibrosis, and expression of adhesion molecules on the endothelium. It also blunted specific parameters of platelet activation and reduced the formation of neutrophil-platelet aggregates, suggesting that dietary ALA may represent a potential therapeutic strategy for SCD.

Summary: This study investigated the optimization of retinal DHA levels through dietary ALA intake. The results showed that a diet with a LA:ALA ratio of 1:1 during pregnancy and assessment of retinal DHA levels in offspring can achieve optimized retinal DHA levels.

Strength training alters the tissue fatty acids profile and slightly improves the thermogenic pathway in the adipose tissue of obese mice

Diego Gomes de Melo et al.

Summary: Short-term strength training was shown to decrease fatty acid levels and lipogenesis enzyme expression in subcutaneous adipose tissue (sWAT) of obese mice. Additionally, it increased the expression of browning-related genes. These effects were observed independently of changes in body weight.

Intranasal delivery of pro-resolving lipid mediators rescues memory and gamma oscillation impairment in AppNL-G-F/NL-G-F mice

Ceren Emre et al.

Summary: In Alzheimer's disease, sustained microglial activation and increased pro-inflammatory signaling lead to chronic inflammation and neuronal damage. This study found that the intranasal administration of pro-resolving lipid mediators improved inflammation, memory deficits, and reduced microglial activation in a mouse model for Alzheimer's disease. These findings suggest a potential future therapy for Alzheimer's disease.

Summary: Chen et al. discovered that the IRE1 alpha endoplasmic reticulum stress sensor inhibits beige adipocyte activation in white and beige adipocytes by regulating lipolysis and Pgc1 alpha messenger RNA levels. The research shows the therapeutic potential of blocking IRE1 alpha in unlocking adipocytes' thermogenic capacity to combat obesity and metabolic disorders.

Discovery of ciliary G protein-coupled receptors regulating pancreatic islet insulin and glucagon secretion

Chien-Ting Wu et al.

Summary: Multiple G protein-coupled receptors (GPCRs) are localized to primary cilia in pancreatic islet cells, with FFAR4 and PTGER4 stimulating cAMP signaling and hormone secretion. The transport of GPCRs to cilia requires TULP3, which plays a key role in regulating hormone secretion by islet cells.

Docosahexaenoic acid ameliorates autoimmune inflammation by activating GPR120 signaling pathway in dendritic cells

Chunlei Feng et al.

Summary: Research shows that omega-3 polyunsaturated fatty acids (n-3 PUFAs), particularly docosahexaenoic acid (DHA), regulate the functional activities of dendritic cells (DCs) to enhance anti-inflammatory effects in autoimmune diseases. By modulating DCs, DHA reduces proinflammatory cytokine production and increases regulatory T-cell induction capability.

Molecular insights into lipoxygenases for biocatalytic synthesis of diverse lipid mediators

Jung-Ung An et al.

Summary: Oxylipins derived from C20- and C22-polyunsaturated fatty acids, known as lipid mediators (LMs), are crucial signaling molecules involved in maintaining homeostasis and promoting healing responses to infection and inflammation in humans. Specialized pro-resolving mediators (SPMs) are a subset of LMs that play a key role in resolving inflammation and infection by activating phagocytosis. Microbial LOXs offer the potential for more efficient synthesis of human LMs from PUFAs, with increased activity and stability compared to human LOXs.
