Summary: The study identified a genetic alteration in vascular cancer epithelioid hemangioendothelioma (EHE) that generates a WWTR1(TAZ)-CAMTA1 (TC) fusion gene. TC is a transcriptional coactivator that drives EHE tumorigenesis. CTGF was identified as a transcriptional target of TC, and its signaling occurs via the Ras-MAPK cascade. Inhibition of MAPK signaling using PD 0325901 and trametinib can abrogate TC-driven tumorigenesis. These findings provided the rationale for a preclinical study evaluating the efficacy of trametinib in patients with unresectable or metastatic EHE.
