Summary: The treatment of chronic myeloid leukemia (CML) has been improved with the development of small-molecule tyrosine kinase inhibitors (TKIs) targeting the fusion protein BCR-ABL1. Ponatinib is a 3rd generation TKI that has high affinity for BCR-ABL1 and is able to inhibit various BCR-ABL1 mutants, including the T315I mutation. Lower starting doses of ponatinib have been recommended by the 2020 European LeukemiaNet for patients with lesser degrees of resistance or multiple intolerances.
