Esterase-Activated Theranostic Prodrug for Dual Organelles-Targeted Imaging and Synergetic Chemo-Photodynamic Cancer Therapy

Jiabao Zhuang et al.

Summary: This report presents a theranostic prodrug, TPE-QC, for dual organelles-targeted and image-guided cancer therapy through synergetic chemotherapy and photodynamic therapy. By reacting with esterase, TPE-QC can be selectively hydrolyzed to release anticancer drug chlorambucil and photosensitizer TPE-QO, resulting in enhanced anticancer potency. The fluorescence of TPE-QC can monitor the release of the prodrug, and the activated TPE-QC accumulates in lysosome and mitochondria, showing efficient tumor therapeutic effects.

Rational Design and Application of an Indolium-Derived Heptamethine Cyanine with Record-Long Second Near-Infrared Emission

Xiaoxie Ma et al.

Summary: In this work, a novel indolium-like heptamethine cyanine dye was designed and developed by installing indolium-derived polycyclic aromatic hydrocarbons on a conjugated polyene backbone. This dye showed bright fluorescent emission in the second near-infrared (NIR-II) window and had the capability of marking blood vessels in mice. Moreover, it exhibited strong absorption bands in the NIR range and high photothermal conversion efficiency, making it suitable for tumor-targeted photothermal therapy.

Dithienylethene-Bridged Fluoroquinolone Derivatives for ImagingGuided Reversible Control of Antibacterial Activity

Haining Zhang et al.

Summary: The emerging field of photopharmacology offers a promising alternative to combat bacterial resistance. However, concerns over degradation, toxicity, and thermal reversibility have hampered its potential applications. In this study, two novel dithienylethene-bridged dual-fluoroquinolone derivatives were developed, displaying efficient photochromism and fluorescence switching behavior. These derivatives also showed improved antibacterial activities compared to monofluoroquinolone derivatives, and demonstrated photoswitchable bacterial imaging.

Growth of Cu2O Nanoparticles on Two-Dimensional Zr-Ferrocene- Metal-Organic Framework Nanosheets for Photothermally Enhanced Chemodynamic Antibacterial Therapy

Xinshuo Zhao et al.

Summary: This study demonstrates the growth of Cu2O nanoparticles on 2D Zr-ferrocene-MOF nanosheets to prepare 2D composites for photothermally enhanced chemodynamic antibacterial therapy. The Cu2O/Zr-Fc-MOF composite shows higher chemodynamic activity than the pristine Zr-Fc-MOF nanosheets, and this activity can be further enhanced by NIR laser irradiation.

Green-/NIR-light-controlled rapid photochromism featuring reversible thermally activated delayed fluorescence and photoelectronic switching

Ziyong Li et al.

Summary: Fluorescent dithienylethene-based photochromic molecule BFBDTE exhibits visible light-controlled fluorescence and thermally activated delayed fluorescence (TADF) switching, as well as rapid and repeatable cyclization and cycloreversion reactions in PMMA. It also demonstrates a remarkable on-off photoelectronic response between the open- and closed-form and holds promise for applications in photoswitchable molecular devices and optical memory storage systems.

Heavy-Atom-Free Photosensitizers: From Molecular Design to Applications in the Photodynamic Therapy of Cancer

Van-Nghia Nguyen et al.

Summary: Photodynamic therapy (PDT) has great potential for cancer treatment due to its spatiotemporal selectivity and noninvasive nature, but the lack of ideal photosensitizers hinders its full potential. Developing advanced smart photosensitizers without heavy atoms has shown significant progress in better meeting the clinical requirements of PDT.
