Manipulation of cadmium and diethylhexyl phthalate on Rana chensinensis tadpoles affects the intestinal microbiota and fatty acid metabolism

Summary: The gastrointestinal tract and intestine microbiota have significant effects on lipid metabolism and immune function in amphibians. Environmental pollution can influence the composition and structure of the microbiota. This study investigated the effects of exposure to Cd and/or DEHP on the morphological indices, histological indices, intestinal microbial community, and mRNA expression levels in Rana chensinensis tadpoles. The results showed that Cd treatment significantly decreased morphological and histological indices, while the mixed treatment group was similar to the control group. Changes in the relative abundance of specific bacterial phyla were observed in the Cd and/or DEHP treatment groups. There were also significant alterations in the mRNA expression levels of lipid metabolism and SCFAs-related genes. These findings suggest that exposure to Cd and/or DEHP can impair lipid storage and transport and reduce anti-inflammatory capacity in amphibians, ultimately leading to a decline in population.

Phthalate-induced testosterone/androgen receptor pathway disorder on spermatogenesis and antagonism of lycopene

Summary: Male infertility is a significant concern due to the decline in sperm quality worldwide. Phthalates, particularly DEHP or its metabolite MEHP, have negative effects on male reproductive development and function, leading to reduced fertility. Lycopene, a natural antioxidant, has the potential to be used as a therapeutic option for male infertility. This research demonstrates that DEHP reduces testosterone production in Leydig cells and impairs secretory function in Sertoli cells, resulting in impaired spermatogenesis. Additionally, MEHP causes mitochondrial and oxidative damage, posing a serious threat to spermatogenesis. However, LYC supplementation reverses these changes. Mechanistically, DEHP disrupts the T/AR signaling pathway, contributing to male infertility. Overall, this study highlights the critical role of the T/AR signal transduction in male fertility and provides promising insights into the protective role of LYC in phthalate-induced male reproductive disorders.
