More contemporary approaches such as read-across, molecular dynamics, and protein modeling are proposed to accelerate the development of toxicity information on PFAS, as well as the need for precaution to protect human health given the known effects of certain PFAS compounds.

Recent US State and Federal Drinking Water Guidelines for Per- and Polyfluoroalkyl Substances

Summary: PFAS are a class of synthetic chemicals that have been produced for over 70 years, with increasing concerns due to their widespread environmental presence, extreme persistence, bioaccumulative nature, and evidence for health effects from environmentally relevant exposures. Multiple US states have found the USEPA Health Advisories insufficiently protective and have developed more stringent guidelines for PFOA, PFOS, and other PFAS, with additional state guidelines expected to be released. The variations in state guidelines for PFAS reflect differences in toxicity and exposure considerations, with the range not unexpected given the different scientific approaches and historical context.

Application of a Framework for Grouping and Mixtures Toxicity Assessment of PFAS: A Closer Examination of Dose-Additivity Approaches
