Residue-Specific High-Resolution 17O Solid-State Nuclear Magnetic Resonance of Peptides: Multidimensional Indirect 1H Detection and Magic-Angle Spinning

Ivan Hung et al.

Summary: This study addresses the challenges of low sensitivity, low resolution, and large quadrupolar couplings in the NMR study of oxygen by employing efficient isotopic labeling, high magnetic fields, fast sample spinning, and H-1 detection. Oxygen sites specific to each amino acid residue are observed through multidimensional experiments. The use of cross-polarization at high sample spinning frequencies enables efficient C-13<->O-17 polarization transfer. Results show that using O-17 for initial polarization provides better sensitivity per unit time compared to H-1.

17O NMR Studies of Yeast Ubiquitin in Aqueous Solution and in the Solid State

Binyang Lin et al.

Summary: A general method for amino acid-type specific O-17-labeling of recombinant proteins in Escherichia coli has been reported. This study demonstrates the first detection of O-17 NMR signals from protein terminal residues and side chains in aqueous solution, as well as solid-state O-17 NMR spectra for labeled proteins obtained at an ultrahigh magnetic field. These findings represent a significant advancement in the field of O-17 NMR studies of proteins.

Looking into the dynamics of molecular crystals of ibuprofen and terephthalic acid using 17O and 2H nuclear magnetic resonance analyses

Chia-Hsin Chen et al.

Summary: This study investigates the structure and dynamics of crystalline ibuprofen and terephthalic acid using O-17 and H-2 NMR analyses, with improved labelling protocols and various experimental methods. The dynamics around carboxylic groups of ibuprofen were studied, revealing motions related to concerted double proton jump and H-bonded unit flip. Preliminary experiments with a H-2-O-17 diplexer connected to the probe were also conducted, showing potential for recording NMR spectra at different temperatures without probe configuration changes.

Expanding the NMR toolkit for biological solids: oxygen-17 enriched Fmoc-amino acids

Brittney A. Klein et al.

Summary: This study reports solid-state O-17 NMR parameters for five previously uncharacterized Fmoc-protected amino acids, achieving efficient O-17 labeling of carboxylic acid moieties through multiple-turnover reactions. Experimental and DFT-computed NMR parameters are in good agreement, supporting the identification of minor unprotonated species. The research highlights O-17 as a sensitive probe nucleus and emphasizes the importance of developing solid-state O-17 NMR techniques for exploring biologically relevant molecules.
