4′-Modified Nucleosides for Antiviral Drug Discovery: Achievements and Perspectives

Summary: Modified nucleosides have therapeutic potential in antiviral treatments, but face new challenges in the era of the COVID-19 pandemic. 4'-Modified nucleosides are an important subclass that can enhance antiviral activity and safety. Several 4'-modified nucleoside drug candidates have progressed into clinical stages, with FNC showing promise in treating HIV-1 and COVID-19.

4′-Fluorouridine is an oral antiviral that blocks respiratory syncytial virus and SARS-CoV-2 replication

Summary: This study describes a drug called 4'-fluorouridine (4'-FlU), which has broad-spectrum antiviral effects and potential for the treatment of respiratory syncytial virus (RSV), severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), and related RNA virus infections. In vitro and in vivo experiments showed that 4'-FlU inhibited RSV, SARS-CoV-2, and related RNA viruses with high efficacy, making it a promising candidate for broad-spectrum therapeutics against respiratory viruses.

Influence of 4′-Substitution on the Activity of Gemcitabine and Its ProTide Against VZV and SARS-CoV-2

Summary: The paper describes the synthesis of novel gemcitabine analogues and their prodrugs, with one analogue showing activity against varicella zoster virus. Despite decreased anti-viral activity upon prodrug derivatization, a concomitant improvement in selectivity index was observed. When tested against SARS-CoV-2, the prodrug exhibited antiviral activity comparable to or slightly lower than its cytotoxic concentration in measurements of cell growth and morphology.

Summary: Fluoro-substitution on the ribose moiety is a common way to modulate the structure and function of nucleic acids, and in this study, the 4'-F-deoxythymidine (T4'-F) was found to maintain the North conformation tendency in duplexes, causing local structural perturbations. Overall, the 4'-F-deoxyribonucleotide structurally resembles the 2'-F-deoxyribonucleotide but imparts less structural perturbation to the duplexes.

When cultured in medium supplemented with fluoride, these bacterial strains produced nucleocidin 1 along with other compounds, including 4'-fluoro-3'-O-beta-glucosylated adenosine 2.
