The unexpected dominance of secondary over primary nucleation

Secondary nucleation is identified as the dominant nucleation mechanism, and enantioselective secondary nucleation can be used to control the chirality of the product, making antisolvent crystallization a viable crystallization-enhanced deracemization technique.

Stochastic shelf-scale modeling framework for the freezing stage in freeze-drying processes

Summary: Freezing and freeze-drying processes are commonly used in pharmaceutical formulations to improve stability. However, batch heterogeneity can cause process failure. In this study, a modeling framework for large-scale freezing processes was developed and an open-source implementation was published. The model couples heat transfer with ice nucleation kinetics and showed how ice nucleation leads to heterogeneity. Various cooling protocols were investigated, and holding schemes were found to have similar solidification times as controlled nucleation, suggesting a potential pathway for process optimization.

Methods to stabilize aqueous supercooling identified by use of an isochoric nucleation detection (INDe) device

Various system modifications were found to significantly enhance the stability of supercooling. Combining Mathematical Modeling and Thermal Infrared Data in the Freezing of Pharmaceutical Liquid Formulations

Summary: In this study, infrared thermal imaging data and mathematical modeling were combined to describe the freezing process of a pharmaceutical formulation during lyophilization. Different mathematical models were used to estimate the pore size distribution of the resulting cake based on the thermal data, and the estimated values were compared to experimental data. The supersaturation model showed the best agreement with the experimental values, while minor discrepancies were observed with the other models. However, these models, when combined with a mathematical model for the primary drying phase, provided satisfactory predictions for the product temperature, resistance to vapor flow, and drying end point.

Isochoric freezing and isochoric supercooling as innovative postharvest technologies for pomegranate preservation

Summary: The present study investigated the effects of isochoric supercooling and freezing on the qualitative attributes of pomegranates, including color, texture, and phytochemical components such as anthocyanins and ascorbic acid. Isochoric supercooling was found to retain color and texture, increase anthocyanins and ascorbic acid content, and inhibit bacterial growth. Isochoric freezing, on the other hand, was the most effective method for inhibiting microbial growth and increasing ascorbic acid content. However, it caused some loss in texture. Isochoric supercooling was also effective in preserving fresh-cut arils, minimizing texture and color loss.

Conceptual Validation of Stochastic and Deterministic Methods To Estimate Crystal Nucleation Rates

Summary: This work presents a generalized framework to assess the accuracy of methods for estimating primary and secondary nucleation rates. Simulation of the crystallization process was conducted using a novel stochastic modeling methodology. It was found that deterministic methods tend to overpredict rates in the presence of secondary nucleation, while stochastic methods were accurate irrespective of secondary nucleation. A criterion was proposed to evaluate the accuracy of stochastic methods. Additionally, the study showed how deterministic and stochastic considerations can be combined to infer both primary and secondary nucleation rates.

Homogeneous freezing of water droplets for different volumes and cooling rates

Summary: To understand ice formation in different environments, it is important to quantify the rate at which ice nucleates from liquid water. Microfluidic platforms, such as the Microfluidic Ice Nuclei Counter Zurich (MINCZ), can provide accurate measurements of nucleation rates. By conducting experiments and simulations, we demonstrate the significance of obtaining a large dataset to improve the accuracy of nucleation rate predictions. Furthermore, we obtain new parameters for a previously developed parameterisation.

Investigation of the Freezing Phenomenon in Vials Using an Infrared Camera

IR thermography was found to be a promising tool for experimentally predicting the resulting product morphology.
