Total Synthesis of Eliglustat via Diastereoselective Amination of Chiral para-Methoxycinnamyl Benzyl Ether

Younggyu Kong et al.

Summary: The total synthesis of eliglustat, an approved drug for non-neurological Gaucher disease, starting from 1,4-benzodioxan-6-carbaldehyde is described. The synthesis involves Sharpless asymmetric dihydroxylation and diastereoselective amination steps, with chlorosulfonyl isocyanate as a key reactant.

Total synthesis of a-1-C-propyl-3,6-di-epi-nojirimycin and polyhydroxyindolizidine alkaloids via regio- and diastereoselective amination of anomeric acetals

Yeon Su Kim et al.

Summary: The synthesis of a-1-C-propyl-3,6-di-epi-nojirimycin and Polyhydroxyindolizidine alkaloids from readily available a-methyl-D-mannopyranoside is described. The key step in this process involves a regioselective and diastereoselective amination of the anomeric acetal functionality using chlorosulfonyl isocyanate (CSI).

Isolated compounds from Dracaena angustifolia Roxb and acarbose synergistically/additively inhibit α-glucosidase and α-amylase: an in vitro study

Jiling Yi et al.

Summary: This study confirmed the potential of Dracaena angustifolia Roxb in regulating glucose dysregulation through in vitro experiments and computer simulations. Several compounds found in Dracaena angustifolia Roxb showed high inhibitory effects on alpha-glucosidase, with synergistic effects observed in combination with acarbose. Additionally, one compound was predicted to be a potential inhibitor of human enzymes. These findings are important for the application of Dracaena angustifolia Roxb in diabetes treatment.

A review on α-glucosidase inhibitory activity of first row transition metal complexes: a futuristic strategy for treatment of type 2 diabetes

Marzieh Sohrabi et al.

Summary: Type 2 diabetes mellitus is a global public health issue, and inhibition of alpha-glucosidase is the main therapeutic approach. Development of non-sugar based inhibitors is gaining attention in addition to research on sugar-based inhibitors. Metal complexes' in vitro anti-alpha-glucosidase activity is attracting increasing attention.

Total Syntheses of Kirkamide and N-acetyl ent-Conduramine B-1

Chintam Narayana et al.

Summary: The second total synthesis of a newly isolated C7N aminocyclitol, kirkamide, was successfully accomplished. Starting from N-acetylglucosamine, the enantiopure epoxide was used to synthesize kirkamide and N-acetyl ent-conduramine B-1 on a gram scale. Key synthetic steps included stereoselective epoxidation, acid-mediated regioselective epoxide ring opening, and selective olefination.
