Improved therapeutic index of an acidic pH-selective antibody

Peter S. Lee et al.

Summary: The researchers hypothesized that an acidic pH-selective ipilimumab may have a better therapeutic index than the traditional ipilimumab. pH Ipi variants have been engineered to have enhanced affinity to CTLA-4 at acidic tumor microenvironment. The experiments conducted on mice showed that these variants maintained anti-tumor activity and reduced peripheral activation, indicating potential for improved safety profiles.

Mitochondrial fission links ECM mechanotransduction to metabolic redox homeostasis and metastatic chemotherapy resistance

Patrizia Romani et al.

Summary: Romani et al. found that cells on soft extracellular matrix have increased mitochondrial fission and production of mitochondrial reactive oxygen species, which may contribute to resistance to chemotherapy drugs in breast cancer cells. The mechanical properties of the tissue also influence the response to treatment. The study suggests that a soft extracellular matrix enhances redox homeostasis and resistance to oxidative stress and chemotherapy drugs.

Glutamine deficiency in solid tumor cells confers resistance to ribosomal RNA synthesis inhibitors

Melvin Pan et al.

Summary: Ribosome biogenesis is regulated by nutrient availability. Nutrient deprivation impairs pre-rRNA processing and leads to the accumulation of unprocessed rRNAs. Restoration of nutrients allows for the processing of stored pre-rRNAs and subsequent ribosome biogenesis. Failure to accumulate pre-rRNAs during nutrient stress disrupts ribosome assembly and activates p53-mediated apoptosis upon nutrient restoration.

Metabolic Codependencies in the Tumor Microenvironment

Prasenjit Dey et al.

Summary: Cancer cell metabolic reprogramming is driven by signals from both intrinsic and extrinsic factors. Intrinsic signaling maintains the baseline metabolic state, while extrinsic signals fine-tune metabolic processes based on metabolite availability and cellular requirements. Therefore, successful targeting of metabolic pathways will require a nuanced approach based on cancer genotype, tumor microenvironment composition, and tissue location.

Cell-programmed nutrient partitioning in the tumour microenvironment

Bradley Reinfeld et al.

Summary: The study found that in the tumor microenvironment, different cell types have varying abilities to access and consume glucose and glutamine, with nutrient partitioning being programmed in a cell-intrinsic manner through mTORC1 signaling and the expression of genes related to glucose and glutamine metabolism.

ORP1L mediated PI(4)P signaling at ER-lysosome-mitochondrion three-way contact contributes to mitochondrial division

Maxime Boutry et al.

Summary: The study reveals that during mitochondrial division, the endoplasmic reticulum recruits lysosomes through the interaction of VAPs with ORP1L, creating a three-way contact between the ER, lysosomes, and mitochondria. Additionally, ORP1L may transport PI(4)P from lysosomes to mitochondria, demonstrating a direct role of PI(4)P in the fission process of mitochondria.
