A Study in Yellow: Investigations in the Stereoselectivity of Ene-Reductases

Fabio Parmeggiani et al.

Summary: Ene-reductases from the Old Yellow Enzyme (OYE) superfamily are efficient biocatalytic alternatives for asymmetric reduction of C=C bonds, offering broad substrate tolerance and excellent stereoselectivities. They can produce a wide range of chiral intermediates used in the synthesis of pharmaceuticals, agrochemicals, flavors, fragrances, and fine chemicals.

One-pot synthesis of dihydro-beta-ionone from carotenoids using carotenoid cleavage dioxygenase and enoate reductase

Zhipeng Qi et al.

Summary: In this study, a new synthesis pathway for dihydro-beta-ionone production was successfully designed and constructed in vitro. By optimizing the reaction conditions and introducing an auxiliary system, a higher yield of dihydro-beta-ionone was achieved. The results of this study demonstrate the feasibility of synthesizing dihydro-beta-ionone from carotenoids and provide guidance for further engineering in the food industry.

Discovery of an ene-reductase for initiating flavone and flavonol catabolism in gut bacteria

Gaohua Yang et al.

Summary: This study identified an ene-reductase, FLR, from the gut bacterium Flavonifractor plautii ATCC 49531 that catalyses the hydrogenation of flavones and flavonols, shedding light on microbial metabolism of flavonoids in the human gut. The crystal structure analysis provided insights into the distinct catalytic property of FLR, which represents a new class of ene-reductases. Genetic and biochemical analyses highlighted the significance of FLR in the gut microbial consumption of dietary and medicinal flavonoids, offering potential implications for personalized nutrition and medicine.

A New Thermophilic Ene-Reductase from the Filamentous Anoxygenic Phototrophic Bacterium Chloroflexus aggregans

Marina Simona Robescu et al.

Summary: The study investigated a new thermophilic ene-reductase homologue, CaOYE, from the photosynthetic extremophile microorganism Chloroflexus aggregans, showing high potential as a robust biocatalyst with high thermostability and good solvent tolerance. Although it displayed a limited substrate spectrum, further investigations could reveal peculiar chemical activities due to the metabolic ductility and versatility of Chloroflexus aggregans. The structural and functional characterization of CaOYE provides new insights for the development and application of this class of enzymes.
