Cross-linked enzyme aggregates immobilization: preparation, characterization, and applications

Summary: Enzyme immobilization, specifically through cross-linked enzyme aggregates (CLEAs), is an effective method to improve enzyme reusability and stability. CLEAs offer several advantages, including high catalytic specificity and selectivity, good operational and storage stabilities, and the ability to co-immobilize different enzymes. This technique finds applications in environmental protection, industrial chemistry, food industry, and pharmaceutical synthesis and delivery.

Application of cellulosic materials as supports for single-step purification and immobilization of a recombinant β-galactosidase via cellulose-binding domain

Summary: This study developed a one-step purification and immobilization method for β-galactosidase on cellulosic supports. The immobilized enzyme showed high activity within 15 minutes and lower sensitivity to galactose compared to the soluble enzyme. Magnesium ion had the highest impact on the enzymatic activity of the immobilized enzyme on cellulosic supports. The derivatives had no cytotoxic effect on cell cultures and retained high hydrolysis capacity after multiple reuse cycles.

8-Galactosidase mediated synthesized nanosupport for the immobilization of same enzyme: Its stability and application in the hydrolysis of lactose

Summary: β-Galactosidase was immobilized on modified nanosilver reduced graphene oxide (Ag@rGO) nanocomposite prepared by in vitro synthesis using same enzyme. The immobilized enzyme showed enhanced activity with an effectiveness factor (eta value) of 0.968, and retained 85% activity after repeated use. The enzyme displayed high activity even in the presence of inhibitory sugars, and effectively hydrolyzed lactose from whey. Additionally, the cytotoxicity and genotoxicity of Ag@rGO nanocomposite was assessed on human blood lymphocytes.
