Intermittent protein restriction protects islet 13 cells and improves glucose homeostasis in diabetic mice

Siying Wei et al.

Summary: This study explored the effectiveness of intermittent protein restriction (IPR) in diabetes control and found that it can significantly reduce hyperglycemia and improve the number and function of pancreatic islet β cells. Furthermore, IPR can reduce glucose production in the liver and enhance insulin signaling in skeletal muscle.

Role of Branched-Chain Amino Acid Metabolism in Type 2 Diabetes, Obesity, Cardiovascular Disease and Non-Alcoholic Fatty Liver Disease

Paola Cuomo et al.

Summary: Branched-chain amino acids (BCAAs) play an important role in human health. High levels of BCAAs are associated with various metabolic diseases, while low levels are beneficial for metabolic health. However, the exact mechanisms underlying the dysregulation caused by high levels of BCAAs are still under debate.

Intermittent Fasting in Weight Loss and Cardiometabolic Risk Reduction: A Randomized Controlled Trial

Sek Ying Chair et al.

Summary: This study investigated the effects of alternate-day fasting (ADF) and 16/8 time-restricted fasting (16/8 TRF) on weight loss, blood glucose, and lipid profile in overweight and obese adults with prediabetes. The findings showed that both ADF and 16/8 TRF were effective in promoting weight loss and improving metabolic markers in this population, with ADF having greater reductions in body weight and mass index compared to 16/8 TRF.

Effect of a plant-based, low-fat diet versus an animal-based, ketogenic diet on ad libitum energy intake

Kevin D. Hall et al.

Summary: In an inpatient, randomized controlled crossover trial, participants consumed 550-700 fewer calories per day when following a plant-based, low-fat diet with a high glycemic load compared with an animal-based, low-carbohydrate diet with a low glycemic load; weight loss was comparable between the two diets and there were no significant differences in hunger or enjoyment of the meals.

Intermittent fasting: from calories to time restriction

Eleonora Duregon et al.

Summary: Calorie restriction (CR) is a dietary manipulation that can increase health and longevity, and recently, the length and periodicity of daily fasting have emerged as potential drivers behind beneficial health effects. Numerous strategies and eating patterns have been successfully developed to recapitulate the benefits of CR.

The adverse metabolic effects of branched-chain amino acids are mediated by isoleucine and valine

Deyang Yu et al.

Summary: The study found that different branched-chain amino acids (BCAAs) have distinct metabolic effects, with low isoleucine diet improving hepatic insulin sensitivity and energy expenditure. It is suggested that reducing isoleucine may be a new approach to treating and preventing obesity and diabetes.

Targeting β-cell dedifferentiation and transdifferentiation: opportunities and challenges

Wenrui Wang et al.

Summary: The review focuses on the pathological characteristics of diabetes mellitus induced by various stressors or immune-mediated injuries, with a particular emphasis on the reduction of pancreatic islet beta-cell populations and activity. Current treatment strategies are ineffective in slowing disease progression, prompting ongoing research in genetically engineering beta-cell mimetics through bi-directional plasticity. The consensus points to beta-cell dedifferentiation as the primary cause of decreased beta-cell mass and activity, suggesting a potential therapeutic window for intervention. Additionally, exploring strategies for beta-cell regeneration using genetic programming, small molecules, cytokines, and bioengineering is discussed, along with the need for further studies to develop efficient reprogramming methods to generate fully functional beta-cells. Further investigations are deemed necessary to transform diabetes into a potentially curable disease.
