Aging-Conferred SIRT7 Decline Inhibits Rosacea-Like Skin Inflammation by Modulating Toll-Like Receptor 2-NF-KB Signaling

Guo Li et al.

Summary: The study reveals that aged individuals and mice are less prone to rosacea, possibly due to reduced sensitivity of their skin to inflammatory stimuli. Downregulation of TLR2 expression in the skin of aged individuals and mice may contribute to this phenomenon. SIRT7 can improve the symptoms of rosacea by modulating the TLR2-NF-kB signaling pathway.

Rosacea, an infectious disease: why rosacea with papulopustules should be considered a demodicosis. A narrative review

F. M. N. Forton

Summary: Rosacea and demodicosis are common facial conditions in dermatology practice. The relationship between the two is controversial. This review discusses the role of Demodex mites in rosacea using three causality assessment systems. The findings suggest that Demodex proliferation is a necessary factor in the development of papulopustular rosacea (PPR), but its occurrence and severity are influenced by multiple co-factors.

A dose-dependent protective effect of n-3 PUFAs in photoaging by promoting collagen production through MAPK pathway in SKH-1 mouse model

Huan Xue et al.

Summary: This study found that n-3 PUFAs can promote collagen synthesis, reduce collagen degradation, inhibit MMP-1 production and abnormal proliferation of keratinocytes induced by UV, and improve the appearance of the skin. These findings suggest the potential clinical prospect of dietary supplementation of n-3 PUFAs in preventing UV-induced skin damage and photoaging.

Mast cell tryptases in allergic inflammation and immediate hypersensitivity

Jonathan J. Lyons et al.

Summary: The dysregulated mast cell-mediated inflammation and activation are linked to various human diseases, and tryptase, a major mast cell granule protein, is commonly used as a biomarker for diagnosing mast cell disorders. However, the mechanistic roles of tryptase in disease pathogenesis are not fully understood. Studies on human tryptase genetics have shown that different genetic composition may impact the structure of tetrameric mature tryptases, leading to clinical phenotypes seen in patients with mast cell-associated disorders. With the advancement of next-generation sequencing, human genetic approaches will play a key role in understanding the functions of tryptase in mast cell disorders and in new therapeutic development.

Angiogenesis in Chronic Inflammatory Skin Disorders

Hyun Ji Lee et al.

Summary: Angiogenesis plays a critical role in chronic inflammatory skin diseases such as psoriasis, atopic dermatitis, and rosacea. Key angiogenetic factors including VEGF, angiopoietins, and various cytokines are involved in the pathogenesis of these skin disorders. Inhibition of angiogenesis may be a promising strategy for treating chronic, inflammatory skin conditions.

Impact of ultraviolet radiation and exposome on rosacea: Key role of photoprotection in optimizing treatment

Daniel Morgado-Carrasco et al.

Summary: The pathophysiology of rosacea involves a complex interaction among genetics, UV light, microorganisms, impaired skin barrier, neuronal and vascular dysfunction, and immune system disruption. UV radiation is reported to play a key role in the development of rosacea, inducing pro-inflammatory, pro-angiogenic, and pro-fibrotic responses. The use of sunscreens containing specific ingredients has been shown to significantly improve symptoms of rosacea, suggesting their potential in mitigating the effects of UV radiation and other environmental triggers.

mTORC1-Mediated Angiogenesis is Required for the Development of Rosacea

Qinqin Peng et al.

Summary: The study found that angiogenesis was enhanced in both rosacea patients and LL37-induced rosacea-like mice, with inhibition of angiogenesis alleviating rosacea-like features. Activation of mTORC1 in endothelial cells was observed in the lesional skin, and inhibiting mTORC1 decreased angiogenesis and blocked rosacea development. In vitro results showed that inhibiting mTORC1 signaling significantly decreased LL37-induced tube formation of human endothelial cells, suggesting that targeting angiogenesis may be a potential therapy for rosacea.

Antimicrobial Peptide LL-37 Drives Rosacea-Like Skin Inflammation in an Manner

Sung-Hyun Yoon et al.

Summary: Rosacea is a chronic inflammatory skin disease characterized by erythema and pustules, and its pathogenesis may be associated with increased levels of antimicrobial peptide LL-37. Research has shown that NLRP3 inflammasome activation plays a crucial role in LL-37-induced skin inflammation and rosacea pathogenesis.
