Systematic Evaluation of Spinal Cord Injury Animal Models in the Field of Biomaterials

Kest Verstappen et al.

Summary: This systematic review aims to identify a list of relevant animal models of spinal cord injury (SCI) by evaluating the confirmation of SCI and animal survival. A total of 149 well-documented mid-thoracic rat models of SCI were identified, which could serve as a starting point for studying biomaterial-based therapies for SCI. The review also discusses the possible bias in SCI animal models, the difficulty in replicating experiments due to poor reporting, and the clinical relevance of the currently used models.

Current Concepts of Neural Stem/Progenitor Cell Therapy for Chronic Spinal Cord Injury

Hidenori Suzuki et al.

Summary: Chronic spinal cord injury is a devastating condition without effective treatment. Clinical studies using neural stem/neural progenitor cells (NSCs/NPCs) are ongoing and show potential for regenerating neural circuits, demyelinating denuded axons, and providing trophic support to endogenous cells.

Regenerative Rehabilitation and Stem Cell Therapy Targeting Chronic Spinal Cord Injury: A Review of Preclinical Studies

Syoichi Tashiro et al.

Summary: Stem cell medicine has shown promise in functional recovery for acute-to-subacute spinal cord injury, but faces challenges in the chronic phase. Researchers are exploring combinatorial treatments to overcome the treatment-resistant state of chronically injured spinal cords. Regenerative rehabilitation, which combines regenerative medicine with rehabilitation, has gained attention for its safety and ability to induce neuronal plasticity. However, there is a lack of comprehensive reviews on regenerative rehabilitation, particularly regarding the use of physical medicine modalities in the chronic phase. Further investigations on combinatorial treatments incorporating stem cell therapy, regenerative rehabilitation, and medication are needed for robust functional recovery in chronic spinal cord injury.

Regulatory Role of Mesenchymal Stem Cells on Secondary Inflammation in Spinal Cord Injury

Qi-Ming Pang et al.

Summary: This review focuses on the immunoregulatory capacity of mesenchymal stem cells (MSCs) in spinal cord injury (SCI) and their potential in tissue repair. The inflammatory cascade plays a crucial role in the secondary injury of SCI, and transplanting MSCs can regulate various immune cells to create a favorable microenvironment for neurological function recovery.

Hydrogels in Spinal Cord Injury Repair: A Review

Zhenshan Lv et al.

Summary: Traffic accidents and falling objects are the main causes of spinal cord injuries (SCIs), seriously affecting the lives and quality of life of patients. Hydrogels, with their biocompatibility and adjustability, serve as structural scaffolds and slow-release carriers in neural tissue engineering, promoting tissue repair. This review discusses the characteristics and advantages of hydrogels as delivery vehicles in SCI therapy and explores their prospects in clinical research for the treatment of SCI.

The ECM: To Scaffold, or Not to Scaffold, That Is the Question

Jonard Corpuz Valdoz et al.

Summary: ECM in tissue engineering plays a crucial role, with scaffold-based and scaffold-free techniques each having their own advantages and limitations. Scaffold-free techniques are suitable for high-throughput cell aggregates preparation, while scaffold-based techniques are more suitable for modeling organotypic models that mimic the native ECM.

Biomaterial-supported MSC transplantation enhances cell-cell communication for spinal cord injury

Bin Lv et al.

Summary: The spinal cord connects the brain to the peripheral nervous system and peripheral tissues through neurons and various types of glial cells. Neurons in the spinal cord process sensory input and control movement. Neurodegenerative disorders and traumatic injuries affecting the spinal cord result in motor deficits and loss of sensory inputs.

Modulation of Both Intrinsic and Extrinsic Factors Additively Promotes Rewiring of Corticospinal Circuits after Spinal Cord Injury

Yuka Nakamura et al.

Summary: Deleting RhoA and RhoC genes to eliminate extrinsic inhibitory pathways, as well as deleting the PTEN gene to enhance intrinsic regenerative response, were found to promote neural circuit rewiring through a combinatorial approach. While this method increased the potential for rewiring, it did not significantly impact axon regrowth across the lesion and motor recovery.

Anisotropic scaffolds for peripheral nerve and spinal cord regeneration

Wen Xue et al.

Summary: Using tissue engineering as an alternative approach, scaffolds with anisotropic structures play a crucial role in guiding neural outgrowth and reconnection for long-gap peripheral nerve and spinal cord injuries. Recent advances in fabrication techniques, such as anisotropic surface patterns and 3D hydrogel scaffolds, show promising in vitro and in vivo effects in orienting axonal and glial cell growth. Challenges and prospects of anisotropic architectures in tissue engineering are also discussed.
