Acute and Chronic Effects of Rifampin on Letermovir Suggest Transporter Inhibition and Induction Contribute to Letermovir Pharmacokinetics

Summary: Rifampin has acute inhibitory and chronic inductive effects that can cause complex drug-drug interactions. This study aimed to separate the effects of rifampin on letermovir disposition and elimination. The results showed increased letermovir exposure with single-dose rifampin plus letermovir, and modestly decreased letermovir exposure with chronic coadministration of rifampin. Letermovir exposure was markedly decreased when administered 24 hours after the last dose of rifampin.

Pregnane X Receptor-4β-Hydroxycholesterol Axis in the Regulation of Overweight- and Obesity-Induced Hypertension

Summary: This study found that activation of PXR can increase blood pressure, while elevated 4-beta hydroxycholesterol in circulation can lower blood pressure. It was also found that 4-beta hydroxycholesterol in the blood is negatively correlated with blood pressure in obese patients and healthy volunteers. This study reveals a new hypertension regulating pathway associated with overweight and obesity.

Activation of pregnane X receptor induces atherogenic lipids and PCSK9 by a SREBP2-mediated mechanism

Summary: This study investigated the role of PXR receptor in regulating atherogenic lipid levels in humans and found that PXR activation induces cholesterol synthesis, leading to elevated LDL and total cholesterol. The PXR-SREBP2 pathway was identified as a novel regulator of cholesterol and PCSK9 synthesis, providing a molecular mechanism for drug- and chemical-induced hypercholesterolemia.
