Circulating tumour DNA reveals genetic traits of patients with intraductal carcinoma of the prostate

Jinge Zhao et al.

Summary: The study found genetic alterations differences between patients with and without intraductal carcinoma of the prostate (IDC-P), with higher proportions of DNA repair pathway and AR pathway alterations in IDC-P carriers. For IDC-P carriers, TP53 mutation was associated with shorter castration-resistant-free survival, and BRCA2 alteration was related to rapid PSA progression in patients receiving abiraterone treatment.

Clinical and Biological Features of Neuroendocrine Prostate Cancer

Yasutaka Yamada et al.

Summary: NEPC can develop de novo but more commonly arises as a result of lineage plasticity, where prostate cancer cells adopt alternative lineage programs to bypass therapy. Genomic analyses from patient biopsies combined with preclinical modeling have further driven tumor proliferation and expression of downstream neuronal and neuroendocrine lineage pathways.

EAU-EANM-ESTRO-ESUR-SIOG Guidelines on Prostate Cancer-2020 Update. Part 1: Screening, Diagnosis, and Local Treatment with Curative Intent

Nicolas Mottet et al.

Summary: The 2020 EAU-EANM-ESTRO-ESUR-SIOG guidelines on screening, diagnosis, and treatment of localized prostate cancer summarize the most recent recommendations for clinical practice, including risk-adapted strategies, medical imaging utilization, biopsy techniques, patient classification, and treatment options. Recommendations include consideration of moderate hypofractionation for intermediate-risk patients and a combination of local treatment with long-term hormonal therapy for cN1 PCa patients.

De novo neuroendocrine transdifferentiation in primary prostate cancer-a phenotype associated with advanced clinico-pathologic features and aggressive outcome

Eman Abdulfatah et al.

Summary: The neuroendocrine transdifferentiation of high-grade prostate cancer (PCA-NT) is characterized by high-grade nuclei, high mitotic activity, reduced PSA expression, high Ki67, and functional inactivation of the RB1 pathway. Most cases present at an advanced stage with adverse radiological findings, metastasis at the time of diagnosis, and high mortality rates. Pathologists should maintain a sensitive threshold for performing immunostains, particularly Ki67 and CyclinD1, in their day-to-day clinical practice due to the prognostic and therapeutic implications of PCA-NT.

Optimizing the diagnosis and management of ductal prostate cancer

Weranja Ranasinghe et al.

Summary: Ductal adenocarcinoma of the prostate is the most common variant histological subtype of prostate cancer, with an aggressive clinical course. Diagnosis and treatment pose challenges, and evidence for treatment is predominantly based on retrospective data.
