The glial cell's role in antinociceptive differential effects of oxytocin upon female and male rats

Ana B. Salinas-Abarca et al.

Summary: Sexual dimorphism plays a significant role in pain processing and response to analgesic drugs. Oxytocin showed a preference for preventing LPS-induced hypersensitivity in male rats compared to females, indicating a potential gender-specific analgesic effect. The results suggest that different strategies may be needed to treat pain in men and women, with oxytocin potentially being more effective for pain with an inflammatory component in men.

Anti-nociceptive effects of ECa 233 a standardized extract of Centella asiatica (L.) Urban on chronic neuropathic orofacial pain in mice

Ananya Buapratoom et al.

Summary: ECa 233 showed anti-nociceptive effects on chronic neuropathic orofacial pain in mice by modulating peripheral calcitonin gen-related peptide expression, which decreased nociceptive activity in trigeminal nucleus caudalis. This study also found a synergistic effect of ECa 233 with gabapentin, reducing mechanical hyperalgesia and allodynia in mice. Further clinical trials are needed to explore the efficacy of ECa 233 in treating neuropathic pain in humans.

Sex-dependent pronociceptive role of spinal α5-GABAA receptor and its epigenetic regulation in neuropathic rodents

Urzula Franco-Enzastiga et al.

Summary: This study examined the sexual dimorphism in neuropathic rodents regarding alpha(5)-GABA(A) receptors, revealing an increase in receptor expression in DRG of nerve-injured females and a decrease in males. Ovariectomy abolished the antiallodynic effect of L-655,708 in females, which was restored by estradiol supplementation. In males, DNA methylation inhibition increased the receptor expression and enabled the antinociceptive effect, suggesting a potential treatment target for chronic pain in females.

CGRP Modulates Orofacial Pain through Mediating Neuron-Glia Crosstalk

H. Liang et al.

Summary: CGRP is upregulated in trigeminal neurons and SGCs following orofacial pain induction. Knockdown of CGRP reduces pain and its expression in neurons and SGCs. CGRP upregulates inducible nitric oxide synthase in SGCs through p38 signaling pathway, leading to a positive-feedback neuron-glia crosstalk mediated by CGRP.

Cannabidiol: pharmacology and therapeutic targets

Stevie C. Britch et al.

Summary: CBD products are commonly used for self-treatment without regulatory approval, with limited scientific evidence from clinical trials. Epidiolex is the only rigorously evaluated product approved for intractable childhood-onset seizures, while studies on CBD for other medical conditions are limited in number and often lack scientific rigor.

Endocannabinoid System Attenuates Oxaliplatin-Induced Peripheral Sensory Neuropathy Through the Activation of CB1 Receptors

Anamaria Falcao Pereira et al.

Summary: This study found that modulation of the endocannabinoid system through the CB1 receptor can alleviate oxaliplatin-induced peripheral sensory neuropathy. Cannabidiol and WIN 55,212-2 can improve mechanical hyperalgesia and c-Fos expression in the spinal cord, while a CB1 antagonist can accelerate cold allodynia response and a CB2 antagonist fails to modulate oxaliplatin-induced nociceptive behavior.

Review of Literatures: Physiology of Orofacial Pain in Dentistry

Nattapon Rotpenpian et al.

Summary: This article provides a summary of the physiology of orofacial pain in dentistry, focusing on the pain pathway, molecular mechanisms of pain pathophysiology, and new insights into classification of orofacial pain related diseases. It also delves into the possible mechanisms of neuropathic orofacial pain, distinguishing it from other types of pain.

Nociceptive receptors are expressed differently in trigeminal nociception after lingual nerve injury and unilateral external carotid artery occlusion in rats

Aree Wanasuntronwong et al.

Summary: The study investigated the changes in nociceptive activity in two animal models of trigeminal neuropathic pain in rats, finding that both unilateral external carotid artery ischemic reperfusion and lingual nerve crush increased nociceptive activity at the trigeminal nucleus caudalis. However, the fEPSPs and expression of nociceptive peripheral afferent neurons differed between the two models.

Acute and Chronic Pain from Facial Skin and Oral Mucosa: Unique Neurobiology and Challenging Treatment

Man-Kyo Chung et al.

Summary: The oral cavity is a portal into the digestive system with unique sensory properties. The oral mucosa needs to be sensitive and selective to differentiate harmful toxins from edible food, but also insensitive to endure noxious irritation. Chronic pain in the oral mucosa is often due to ulcers, and conditions like atypical odontalgia and burning mouth syndrome pose challenges in diagnosis and treatment.

Evolution of mirror-image pain in temporomandibular joint osteoarthritis mouse model

Nattapon Rotpenpian et al.

Summary: By studying mirror-image pain in an osteoarthritis mouse model of the temporomandibular joint, it was found that nocifensive behaviors on the contralateral side became stronger on day 28, in correlation with elevated phosphorylated cyclic AMP response element binding protein and microglia marker in the trigeminal subnucleus caudalis, indicating the occurrence of mirror-image pain.

A standardized extract of Centella asiatica (ECa 233) prevents temporomandibular joint osteoarthritis by modulating the expression of local inflammatory mediators in mice

Nattapon Rotpenpian et al.

Summary: The study showed that the expression of TRPV1, ASIC3, and CGRP significantly increased in mice with TMJ-OA on day 21 after injecting complete Freund's adjuvant. However, ECa 233 was more effective than ibuprofen in reducing the expression of TRPV1 and ASIC3, thereby inhibiting the pathogenesis of temporomandibular joint osteoarthritis.
