Discovery of a potent Guanidine derivative that selectively binds and stabilizes the human BCL-2 G-quadruplex DNA and downregulates the transcription

Summary: G-quadruplex (G4) structures are non-canonical DNA or RNA secondary structures that can develop in guanine-rich sequences. They have been found in various locations in the human genome and are recognized as potential targets for cancer therapeutics. In this study, a small molecule called GD-1 was found to selectively interact with the BCL-2 GQ structure with high affinity, leading to an increase in its thermostability. Biological experiments showed that GD-1 suppresses the expression of BCL-2 genes in HeLa cells and has potential as an anti-cancer agent.

Preferential stabilization of c-MYC and BCL-2 promoter G-quadruplexes by a natural betaine-type alkaloid

Summary: This study demonstrates that the alkaloid Lycobetaine (LYC) preferentially binds to c-MYC and BCL-2 promoter G-quadruplexes (G4s) and stabilizes them through end-stacking mode. LYC inhibits the expression of c-MYC and BCL-2 oncogenes, leading to cell cycle arrest, apoptosis, as well as inhibition of cell proliferation and migration. These results reveal a new potential anticancer mechanism and application of LYC in drug-resistant cancers.

Piperine analogs arrest c-myc gene leading to downregulation of transcription for targeting cancer

Summary: In this study, Piperine analog PIP-2 was found to have high toxicity against A549 lung cancer cells but low toxicity towards normal cells, indicating its potential as an anticancer therapeutic at the cellular level. Through interaction with cancer cells, PIP-2 was shown to stabilize the G-quadruplex motif at the promoter site of the c-myc oncogene, resulting in downregulation of its expression.
