Summary: Chromosomal ploidy manipulation is an effective method to create excellent germplasm. Triploid fish can serve as a sterile model for studying abnormality in meiosis. High-throughput sequencing was used to compare the expression profiles of gonadal RNA in diploid and triploid female rainbow trout at three developmental stages, identifying a majority of differentially expressed RNAs and characterizing 22 DE mRNAs related to oocyte meiosis and homologous recombination. Predicted miRNA-mRNA/lncRNA networks were constructed to show potential interactions between coding and noncoding RNAs during gonadal development of polyploid fish.

Novel variants in DNAH9 lead to nonsyndromic severe asthenozoospermia

Summary: This study identified DNAH9 as a novel pathogenic gene for nonsyndromic severe asthenospermia using whole exome sequencing and Sanger sequencing. Further experiments showed that in cases of nonsyndromic severe asthenospermia, DNAH9 expression was significantly decreased and intracytoplasmic sperm injection (ICSI) can lead to favorable pregnancy outcomes for these patients.

Summary: In sexually reproducing animals, a subset of RNAs expressed during early oogenesis undergo regulated degradation, failure to remove these RNAs results in oocyte dysfunction and death. Key regulators of oogenesis through targeted degradation of specific RNAs include the RNA-degrading Super Killer complex and No-Go Decay factor Pelota. Targeted RNAs are enriched for cytidine sequences bound by the polypyrimidine tract binding protein Half pint, orchestrating a germ cell-to-maternal transition giving rise to the maternal contribution to the zygote.
