DNA/protein binding and anticancer activity of ruthenium (II) arene complexes based on quinoline dipyrrin

Summary: Two arene ruthenium complexes ([(eta(6)-arene)Ru(L)Cl]) with different arene ligands (benzene, R1; p-cymene, R2) and a chelating ligand (2-chloro-3-(di(1H-pyrrol-2-yl)methyl)quinoline) (L) have been synthesized and characterized. The complexes showed appreciable binding with calf thymus DNA (CT-DNA) and strong binding with bovine serum albumin. DFT studies further supported the intercalative interaction of the complexes with DNA. Both complexes exhibited potential anticancer activity against T cell lymphoma, with R2 showing better activity than R1 (IC50, R1: 30 μM, R2: 20 μM).

The crystal structure analysis revealed a distorted octahedral coordination geometry and various intermolecular interactions. The calculated results using density functional theory (DFT) showed good agreement with the experimental data. The complex exhibited antimicrobial, antioxidant, and anti-inflammatory activities.

Summary: In this study, we synthesized and evaluated the biological activity of new thiosemicarbazones and their cyclized heterocyclic compounds through in silico and in vitro examinations. Two pyrene-based thiosemicarbazone compounds were prepared and two new compounds were obtained through cyclization reactions. The structure of the compounds was characterized using various analytical techniques. Among them, PP2 exhibited strong cytotoxicity and inhibited the growth of HepG-2 and T24 cells.

Interaction of new tri-, tetra-, and pentacyclic azaphenothiazine derivatives with calf thymus DNA: Spectroscopic and molecular docking studies

Summary: In this study, the interaction mode of selected azaphenothiazines (AZA) with calf thymus DNA was investigated using UV-Vis absorption, fluorescence spectroscopy, and molecular docking. The results showed that AZA bind to DNA by inserting a fragment of the fused rings system between the base pair stack in the double helix. The number of rings in the AZA structures seemed to be correlated with the strength of the interaction, with pentacyclic AZA demonstrating higher affinity towards DNA.
