An Efficient Protein Evolution Workflow for the Improvement of Bacterial PET Hydrolyzing Enzymes

Valentina Pirillo et al.

Summary: This study reports a straightforward workflow for the evolution of PET-hydrolysing enzymes (PHEs) using semi-rational protein engineering and high-throughput screening. By screening variant libraries on PET nanoparticles, a variant with higher hydrolytic activity was identified and further stabilized. This workflow provides a suitable protocol for the generation of efficient enzymatic tools for polyester degradation.

Real-Time Noninvasive Analysis of Biocatalytic PET Degradation

Ronny Frank et al.

Summary: The Earth has entered the Anthropocene, characterized by widespread environmental pollution from plastics like PET. There is a pressing need for ecofriendly and cost-effective solutions for plastic recycling and reuse. By altering the dielectric properties of PET films, a monitoring system has been developed using impedance spectroscopy to analyze the enzymatic hydrolysis process of PET films with polyester hydrolases.

Engineering Transcription Factor XylS for Sensing Phthalic Acid and Terephthalic Acid: An Application for Enzyme Evolution

Jiawei Li et al.

Summary: Researchers have developed two novel variants of a transcription factor that can detect PA and TPA and activate the expression of a fluorescent protein. Using these variants, they constructed whole-cell biosensors that can detect as little as 10 μM of PA or TPA. They also engineered a PAE hydrolase to enhance its activity towards DBP and p-nitrophenyl butyrate.

Structural basis of terephthalate recognition by solute binding protein TphC

Trishnamoni Gautom et al.

Summary: The study demonstrates that the presence of the gene encoding the solute binding protein TphC enables the uptake of terephthalate (TPA), allowing for the biological degradation of Polyethylene terephthalate (PET) plastic. The analysis reveals the narrow ligand specificity of TphC towards aromatic para-substituted dicarboxylates, such as TPA.

Potential Use of Microbial Enzymes for the Conversion of Plastic Waste Into Value-Added Products: A Viable Solution

Muhammad Tamoor et al.

Summary: The widespread use of commercial polymers composed of a mixture of polylactic acid and polyethene terephthalate (PLA-PET) in bottles and other packaging materials has caused a massive environmental crisis. The enzymatic hydrolysis of PLA-PET contaminants plays a vital role in environmentally friendly strategies for plastic waste recycling and degradation. Various enzymes and strategies for the utilization of PLA-PET's carbon content as C1 building blocks for the production of new plastic monomers and value-added products were discussed in detail.

Towards bio-upcycling of polyethylene terephthalate

Till Tiso et al.

Summary: With over 359 million tons of plastics produced worldwide in 2018, the challenge of managing plastic waste is evident. Recent discovery of enzymes that degrade supposedly non-biodegradable plastics has opened up opportunities for transforming the plastic recycling industry using biotechnology. This study presents a novel method for converting post-consumer PET into bioplastics, offering a solution that bypasses the expensive purification process for PET monomers and adds technological flexibility to the global issue of plastic waste management.
